摘 要:目的探讨miR-181b在动脉粥样硬化血管中的表达及miR-181b对H 2O2诱导的血管内皮细胞增殖与凋亡的影响。方法 qPCR法检测动脉粥样硬化模型组小鼠主动脉和H2O2诱导的血管内皮细胞中miR-181b的表达情况;酶联免疫法检测miR-181b对鼠血清中TNF-α、IL-6和IL-10含量的影响;克隆形成实验检测miR-181b对H2O2诱导的血管内皮细胞增殖能力的影响;流式细胞术检测miR-181b对H2O2诱导的血管内皮细胞凋亡能力的影响;qPCR和Western blot法检测miR-181b对主动脉和H2O2诱导的血管内皮细胞中Caspase-3、Bax和Bcl-2mRNA和蛋白表达的影响。结果 miR-181b在动脉粥样硬化模型组小鼠主动脉和H2O2诱导的血管内皮细胞表达水平显著下调,血清中TNF-α、IL-6含量均增加,IL-10含量减少;过表达miR-181b使小鼠血清中TNF-α、IL-6含量均减少(P<0.05),IL-10含量增加(P<0.05)。与对照组比较,H2O2诱导的血管内皮细胞的增殖能力显著下降,凋亡能力显著提高,miR-181b过表达使血管内皮细胞增殖能力提高,凋亡能力下降;与对照组比较,动脉粥样硬化模型组小鼠主动脉和H2O2诱导的血管内皮细胞中Caspase-3、Bax的mRNA和蛋白表达水平均上调,Bcl-2mRNA和蛋白表达水平下调,miR-181b过表达使动脉粥样硬化模型组小鼠主动脉和H2O2诱导的血管内皮细胞中Caspase-3、Bax的mRNA和蛋白表达水平均下调,Bcl-2 mRNA和蛋白表达上调(P<0.05)。结论 miR-181b在动脉粥样硬化血管和H2O2诱导的血管内皮细胞中低表达,过表达miR-181b抑制细胞凋亡及促进细胞的增殖,可能在动脉粥样硬化的治疗中起到积极作用。Objective To investigate the expression of miR-181 b in atherosclerotic vessels and the effect of miR-181b on vascular endothelial cell proliferation and apoptosis induced by H2O2. Methods qPCR was used to detect the expression of miR-181 b in aortic atherosclerosis model group mouse aorta and vascular endothelial cells induced by H2O2. The effect of miR-181 b on the levels of TNF-α, IL-6 and IL-10 in serum was detected by ELISA. Cloning assay was used to detect the effect of miR-181 b on the proliferation of vascular endothelial cells induced by H2O2. Flow cytometry was used to detect the effect of miR-181b on the apoptosis of vascular endothelial cells induced by H2O2. qPCR and Western blot were used to detected the effects of miR-181 b on the expression of Caspase-3, Bax and Bcl-2 mRNA and protein in aorta and vascular endothelial cells induced by H2O2. Results miR-181 b was significantly down-regulated in aortic atherosclerosis model mouse aorta and H2O2-induced vascular endothelial cells. Compared with the control group, the levels of TNF-α and IL-6 in serum were increased in the atherosclerosis model group and the level of IL-10 in serum was decreased, overexpression of miR-181 b decreased the levels of TNF-α and IL-6, and increased IL-10. Compared with the control group, the proliferative capacity of vascular endothelial cells induced by H2O2 was significantly decreased, and the apoptotic ability was significantly increased, overexpression of miR-181 b increased the proliferation of vascular endothelial cells and decreased the apoptotic ability. Compared with the control group, the expression levels of Caspase-3 and Bax mRNA and protein were up-regulated in the aorta of the atherosclerosis model and vascular endothelial cells induced by H2O2, and the expression level of Bcl-2 mRNA and protein was down-regulated, overexpressed miR-181b down-regulated the expression of Caspase-3 and Bax mRNA and protein in the aorta of the atherosclerosis model and vascular endothelial cells induced by H2O2, and up-regula
