摘  要:目的探究连翘酯苷A(FTA)对小鼠压力负荷性心力衰竭(HF)的保护作用及其潜在的机制。方法 30只健康雄性C57BL/6小鼠随机分成Sham组、HF组和HF+FTA组。采用主动脉弓缩窄建立小鼠压力负荷性HF。称取小鼠体重及心脏重量并计算心脏重量指数;HE染色观察组织病理学改变;ELISA检测小鼠血清内炎症因子改变;Western blot技术检测相关蛋白表达。结果 FTA显著降低压力负荷性HF引起的小鼠心脏重量和心脏重量指数增加,减少心肌间质炎症细胞浸润,降低血清内炎症因子TNF-α、IL-6和IL-1β表达水平;心肌Nrf2和HO-1蛋白表达增加,NF-κB蛋白表达降低。结论 FTA对小鼠压力负荷性HF具有保护作用,与激活Nrf2/HO-1信号通路相关。Objective To investigate the protective effect of forsythiaside A(FTA) on pressure overload heart failure(HF) in mice. Methods Thirty healthy male C57BL/6 mice were randomly divided into three groups: Sham group, heart failure group(HF) and HF+FTA group. Aortic arch constriction(TAC) was used to establish pressure-loaded HF in mice. The body weight and heart weight was determined to calculate heart weight index;HE staining to observe histopathological changes;ELISA to detect the changes of inflammatory factors in serum of mice;Western blot to detect protein expression. Results Compared with the HF group, FTA treatment significantly reduced mouse heart weight and heart weight index, reduced myocardial interstitial inflammatory cell infiltration and the levels of TNF-α and IL-6 and IL-1β in serum, decreased expression level of NF-κB protein, but increased Nrf2 and HO-1 proteins levels in cardiocytes. Conclusion FTA has a protective effect on pressure overload HF in mice, which is related to activation of the Nrf2/HO-1 signaling pathway.
