摘 要:目的探索慢性苯中毒患者和健康对照人群中lncRNA的差异表达,初步建立慢性苯中毒lncRNA异常表达基因谱。方法采用(4×180 K)lncRNA芯片,对慢性苯中毒患者及健康对照人群外周静脉血各6例共12个标本进行lncRNAs及mRNAs检测,最后采用GO分析和KEGG分析差异表达基因初步生物学功能。结果lncRNA差异表达基因有4806个,其中2432个表达上调,2374个表达下调;mRNA差异表达基因有4131个,其中2281个表达上调,1850个表达下调。GO分析表明,差异表达基因功能注释主要包括有炎症反应、凝血功能、细胞膜有机成分等。KEGG分析表明,差异表达基因主要富集在细胞因子与细胞因子受体交互作用通路、造血细胞系等通路上。结论差异表达的lncRNA为深入研究慢性苯中毒机制提供理论依据。Objective To explore the differential expression of lncRNAs and mRNAs in chronic benzene poisoning and normal control.Methods LncRNA Microarrays(4×180 K)were used to detect discrepant lncRNAs and mRNAs in peripheral lymphocytes from six chronic benzene-poisoned patients and six normal controls.Finally,GO and KEGG were used to analyze the primary biological functions of differential expression genes.Results There were 4806 differential expression lncRNAs between chronic benzene poisoning and normal controls,2432 lncRNAs were upregulated and 2374 were downregulated.In addition,There were 4131 differential expression mRNAs between chronic benzene poisoning and normal controls,2281 mRNAs were upregulated and 1850 mRNAs were downregulated.GO analysis showed that the functional annotation of differential expression genes mainly include inflammatory response,coagulation function and organic components of cell membrane.The interaction pathways of cytokines and cytokines receptors,hematopoietic cell lines and other pathways were showed by KEGG.Conclusion Differentially expressed lncRNAs provide a theoretical basis for further study of the mechanism of chronic benzene poisoning.
