摘  要:目的初步分析20例HBV(Hepatitis B virus,HBV)疫苗志愿接种者(汉族、布依族、苗族、侗族)接种前、后外周血中淋巴细胞(Peripheral blood lymphocyte,PBL)CD4+T细胞受体(T cell receptor,TCR)β链26个TRBV家族互补决定区3(Complementarity determining region3,CDR3)的变化及特征。方法筛选乙肝五项标记物阴性的20例志愿接种者,于0、1、6月进行HBV疫苗接种,PCR扩增并分析志愿接种者接种前、后外周血CD4+T细胞TCRβ链的各家族CDR3谱系漂移的特征。结果20例HBV疫苗志愿接种者在接种后都表现出一个或几个TRBV家族的优势利用,个体乙肝疫苗接种后优势TRBV家族的取用与民族属性表现出具有一定的关联性,而与HLA-DR位点未表现出明显关联性。结论HBV疫苗接种诱导了机体相应的T细胞应答;个体疫苗接种后CD4+T细胞表现出优势利用TRBV家族CDR3的多样性,这种现象可能与TCR CDR3区本身的空间构型和结构、HBV抗原肽的不同抗原表位及HLA型别差异有一定关联性;同时,T细胞对HBV的应答和民族属性表现出一定的关联性。Objective To preliminarily analyze the changes and characteristics of the lymphocytes(PBL)CD4+T cell receptor(T cell receptor,TCR)complementary determination region3(CDR3)of the beta chain in 20 cases of volunteers(Buyi,Dong,Miao,Han)before and after vaccinated against HBV(Hepatitis Bvirus,HBV).Methods Twenty volunteers with negative markers were screened and vaccinated against HBV in the months of 0,1 and 6 After collecting peripheral blood samples,expanding and analyzing the characteristics of CDR3 of 26 TRBV families in pre-inoculation and CD4+T cell TCRβchains of HBV vaccine volunteers.Results The advantages of one or more TRBV family were observed in 20 cases of HBV vaccine volunteers after inoculation.The use of the TRBV family after vaccinated against HBV in different individuals showed a certain correlation with ethnic attributes,but did not show a significant correlation with the HLA-DR.Conclusion HBV vaccination could induce the corresponding T cell response in the body.The advantages of CD4+T cells take advantage of the diversity of the TRBV family CDR3 after vaccinated,the reasons for which are related to the different antigenic sites of HBV antigenic peptides,the HLA type difference,and the structure and spatial configuration of the TCRCDR3 region.At the same time,the response of T cells to HBV shows a certain correlation with ethnic attributes.
