摘 要:目的:建立黄芩药材和饮片的HPLC指纹图谱。方法:采用安捷伦zorbax XDBC18(4.6 mm×250 mm,5μm)色谱柱,流动相为甲醇-水(含0.1%磷酸),梯度洗脱,流速设定为1 m L/min,柱温设定为30℃,进样体积10μL,检测波长为270 nm。结果:分别建立了黄芩药材和饮片的高效液相色谱指纹图谱共有模式,标定了17个共有峰,指认了5个共有峰,分别为汉黄芩素、黄芩苷、黄芩素、野黄芩苷、汉黄芩苷。10批药材相似度不低于0.991,10批饮片相似度均值为0.993。结论:该方法准确、可靠,为黄芩药材与饮片质量标准提升提供数据支持。Objective:To develop and validate the fingerprint analysis method of Radix Scutellariae and its processed pieces by HPLC.Methods:The chromatographic fingerprint was obtained on Agilent zorbax XDB C 18(4.6 mm×250 mm,5μm)and used methanol and water(containing 0.1%phosphoric acid)as mobile phase with the flow rate at 1 mL/min and 10μL of injection volume.The optimal column temperature was 30℃.The detection wavelength was optimized at 270 nm.Results:The common mode of HPLC fingerprint of Radix Scutellariae and its processed pieces was set up.Seventeen common peaks in the fingerprints were found.Scutellarin,baicalin,wogonin,wogonoside,and baicalein were identified.The similarities of 10 batches of Radix Scutellariae were not less than 0.999.The average similarity between 10 batches of processed Radix Scutellariae pieces and the reference fingerprint was 0.993.Conclusion:The fingerprint method was accurate and reliable for Radix Scutellariae and its processed pieces analysis,which can provide data support towards quality standard improvement.
