摘 要:采用免疫磁珠净化法对小麦、玉米等样品进行样本的前处理分析研究,完成多种样品基质的净化和富集,随之利用高效液相色谱法对样品中的黄曲霉毒素B 1含量进行检测分析。为了验证免疫磁珠法快速提取的可行性,在相同的实验条件下采用免疫亲和柱完成对样品提取,对比两种净化方式对样品检测结果的差异性,结果表明两种提取方式的检测结果误差在可接受的范围内。采用免疫磁珠净化法对添加AFB 1的空白小麦、花生、饲料、辣椒酱等样品进行加标检测,平均回收率在88.58%~107.61%,变异系数小于10%,表明免疫磁珠法可规模化地应用于食品安全检测领域。In this study,various sample substrates(wheat,corn,feed,etc.)were pretreated and enriched by the immunomagnetic beads purification method and immunoaffinity column method,respectively.In addition,the content of different samples were detected by the high performance liquid chromatography.The different detection results of the two methods were conducted to determine the feasibility of the immunomagnetic bead method,and the accuacy of two methods was within the error range.The results showed that the average recovery rate was 88.58%-107.61%,and the relative variation coefficient was less than 10%,which indicated that immunomagnetic beads purification method could be applied in food safety field.
