摘  要:为探讨我国2个地方蛋鸡品种仙居鸡和白耳黄鸡遗传多样性和起源进化,以30只仙居鸡和30只白耳黄鸡为素材,通过PCR扩增和测序技术,对其进行线粒体DNA控制区全序列单倍型和遗传多样性分析,并结合GenBank公布的19条红色原鸡D-loop区全序列,构建系统发育树。结果表明:2个鸡品种变异区集中在第167—1 215 bp之间,共发现19个多态位点和9个单倍型,其中仙居鸡和白耳黄鸡各有3种特异单倍型。系统发育分析显示,9种单倍型可分为A、B和C共3个分支,白耳黄鸡的优势单倍型属于B分支,仙居鸡的优势单倍型属于C分支。聚类分析显示,白耳黄鸡主要与原鸡滇南亚种聚为一类,仙居鸡与原鸡印度亚种、滇南亚种、指名亚种和印尼亚种聚为一类,具有多个母系起源。这一研究从分子水平为2个蛋用型地方鸡品种仙居鸡和白耳黄鸡遗传资源开发利用和保护提供了理论依据。To explore the genetic diversity and origin evolution of Xianju chicken and Baier Yellow chicken of our country, 30 Xianju chicken and 30 Baier Yellow chicken were used as material, by PCR amplification and sequencing technologies, haploid type and genetic diversity of all samples on mitochondrial DNA complete sequence were analyzed, and compared to the 19 red jungle fowl sequences in GenBank. The results showed that there were 19 polymorphic sites and 9 haplotypes between 167 bp and 1 215 bp, among which Xianju chicken and Baier Yellow chicken each had 3 specific haplotypes. Phylogenetic analysis showed that the nine haplotypes could be divided into three branches: clade A, clade B and clade C. The dominant haplotypes of Baier Yellow chicken were mainly clustered with clade B, and the dominant haplotypes of Xianju chicken were mainly clustered with clade C. Cluster analysis showed that Baier Yellow chicken was clustered with Gallus gallus spadiceus, Xianju chicken might have multiple origins, and was clustered with Gallus gallus murgha, Gallus gallus spadiceus, Gallus gallus gallus, Gallus gallus bankiva. The results would provide a theoretical basis for genetic resources conservation and development and utilization of Xianju chicken and Baier Yellow chicken.
