摘 要:以新农1号狗牙根种子为材料,在0.0%和15.0%PEG下筛选适宜的茉莉酸甲酯(MeJA)浸种浓度;在此基础上,研究最适MeJA浸种对不同渗透胁迫(PEG-6000质量比依次为0.0%、7.5%、15.0%、22.5%)下新农1号狗牙根种子萌发、幼苗生长、超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量的影响。结果表明,在0.0%和15.0%PEG胁迫下,随MeJA浸种浓度增加,新农1号狗牙根种子发芽率、发芽势、发芽指数及活力指数均呈先升后降趋势。其中,0.15μmol/L MeJA浸种处理的上述发芽指标均达最高,可作为浸种最适浓度。不同浓度PEG渗透胁迫下,与蒸馏水浸种相比,0.15μmol/L MeJA浸种处理可显著增加种苗SOD、CAT活性,降低MDA含量,促进狗牙根种子的发芽指标提升。特别是在高浓度PEG(22.5%)下,缓解渗透胁迫效应更加显著。总之,适宜浓度的MeJA浸种可缓解渗透胁迫对狗牙根种子萌发及幼苗生长的伤害,增强种子萌发期抗旱能力。Using seeds of bermudagrass(Cynodon dactylon L.)‘Xinnong No.1’as tested materials,the appropriate soaking concentration of methyl jasmonate(MeJA)under 0.0%and 15.0%PEG was determined in the present study,in which the effects of MeJA soaking on seed germination,seedling growth,superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)activities and malondialdehyde(MDA)content under different osmosis stress(PEG-6000 mass ratio of 0.0%,7.5%,15.0%and 22.5%)were studied.The results showed that with the increase of MeJA concentration,the germination rate,germination potential,germination index and vigor index of‘Xinnong No.1’increased first and then decreased under 0.0%and 15.0%PEG stress.Especially under osmosis stress,the above germination indexes were the highest in 0.15μmol/L MeJA soaking treatment,which could be considered as the optimal concentration for soaking seeds.Compared with soaking seeds in distilled water,soaking seeds in 0.15μmol/L MeJA significantly increased the activities of SOD and CAT,decreased the content of MDA,and promoted the germination indexes of‘Xinnong No.1’seeds under different concentrations of PEG osmotic stress.Especially under high concentration of PEG(22.5%),the effect of alleviating osmotic stress was more significant.In conclusion,soaking seeds with appropriate concentration of MeJA can alleviate the damage of osmosis stress on seed germination and seedling growth of‘Xinnong No.1’seeds,enhance the drought resistance of seeds during germination stage,and provide technical support for seed establishment of‘Xinnong No.1’under drought conditions.
