摘 要:目的:通过网络药理学研究参地补肾胶囊抗肾间质纤维化(RIF)的物质基础及作用机制。方法:借助TCMSP等数据库检索参地补肾胶囊的化学成分和作用靶点。通过Gene Cards,OMIN生物数据库筛选出RIF的疾病靶点。使用Cytoscape软件构建“药物-成分-靶点-疾病”网络关系,构建蛋白-蛋白相互作用网络,进行基因本体富集分析和京都基因与基因组百科全书通路富集分析。结果:获得参地补肾胶囊286个有效化学成分和抗RIF潜在作用靶点121个,网络分析结果表明参地补肾胶囊主要通过STAT3、TP53、JUN、AKT1、TNF、SRC、MAPK1、MAPK8、IL-6、MAPK14、RELA、VEGFA、EGFR、FOS、ESR1等关键靶点,以及调控流体剪切应力与动脉粥样硬化,HIF-1信号通路、TNF信号通路、VEGF信号通路、ErbB信号通路、Jak-STAT信号通路、NF-κB信号通路等途径发挥抗RIF的作用;尤其HIF-1信号通路、TNF通路、JAK/STAT通路和ErbB信号通路的蛋白和下游蛋白表达常常是其他通路参与RIF的启动因子或其他通路中的关键因子。结论:参地补肾胶囊通过多条通路、多个靶点的综合调控达到抗RIF,治疗CRF的作用;其中HIF-1信号通路、TNF通路、JAK/STAT通路和ErbB信号通路是关键通络。Objective:To study the material basis and mechanism of Shendi Bushen Capsule against renal interstitial fibrosis(RIF)through network pharmacology.Method:The chemical components and target of Shendi Bushen capsule were searched by TCMSP and other databases.The disease targets of RIF were screened out by Gene Cards and OMIN biological database.Use Cytoscape software to build a"drug-component-target-disease"network relationship and a protein-protein interaction network,construct the protein-protein interaction network,and the enrichment analysis of gene ontology and Kyoto gene and genomic encyclopedia pathway was carried out.Results:286 effective chemical components and 121 potential anti-RIF targets of Shendi Bushen Capsule were obtained.The results of network analysis showed that Shendi Bushen Capsule mainly passed STAT3,TP53,JUN,AKT1,TNF,SRC,MAPK1,MAPK8,IL6,MAPK14,RELA,VEGFA,EGFR,FOS,ESR1 and other key targets,as well as regulating the fluid shear stress and atherosclerosis,HIF-1 signaling pathway,TNF signaling pathway,VEGF signaling pathway,ErbB signaling pathway,Jak-STAT signaling pathway,NF-κB signaling pathway and other pathways play an anti-RIF role.Especially the HIF-1 signaling pathway,TNF pathway,JAK/STAT pathway and ErbB signaling pathway.The expression of proteins and downstream proteins in these pathways are often the initiation factors of other pathways participating in RIF or the key factor in other pathways.Conclusion:Shendi Bushen Capsule achieves anti-renal interstitial fibrosis and treatment of CRF through multiple pathways,multiple targets and comprehensive regulation.In particular,HIF-1 signaling pathway,TNF pathway,JAK/STAT pathway and ErbB signaling pathway are the key channels.
