化纤胶囊2号通过调控氧化应激缓解脓毒症诱导的大鼠心肌损伤

摘　　要：目的:通过观察氧化应激作用探讨化纤胶囊2号对脓毒症诱导的大鼠心肌损伤的保护作用。方法:40只大鼠随机分为对照组8只;脓毒症组(CLP)16只,其中12、24 h每组各8只;脓毒症+化纤胶囊2号组16只,其中12、24 h每组各8只。采用盲肠结扎穿刺(CLP)方法诱导大鼠脓毒症,并在术后给予化纤胶囊2号(10 mL/kg)。测定脓毒症诱导的心肌损伤大鼠血清中白介素-1前体(IL-1β)、肿瘤坏死因子-α(TNF-α)的含量,血清中乳酸脱氢酶(LDH)和磷酸肌酸激酶(CK-MB)心肌酶水平。测定化纤胶囊2号对脓毒症诱导的心肌损伤大鼠心肌组织中氧化应激相关指标丙二醛(MDA)水平、谷胱甘肽过氧化物酶(GPx)和超氧化物歧化酶(SOD)活性的水平。结果:化纤胶囊2号可降低脓毒症诱导的心肌损伤大鼠血清中IL-1β、TNF-α的炎症因子水平、降低血清中LDH和CK-MB心肌酶水平;与CLP组相比,治疗组抑制心肌组织的氧化应激增强,增加抗氧化水平;差异均有统计学意义(P<0.01)。结论:化纤胶囊2号减轻脓毒症诱导的心肌损伤,降低脓毒症诱导的心肌损伤大鼠氧化应激水平。Objective:To observe the protective effect of Huaxian CapsuleⅡon sepsis-induced myocardial injury by regulating oxidative stress in rats.Methods:40 rats were randomly divided into the normal control group(n=8),the sepsis group(n=16),which included 8 rats at 12 h and 8 rats at 24 h,and the treatment group(Huaxian CapsuleⅡ,n=16),which also included 8 rats at 12 h and 8 rats at 24 h.Sepsis was induced in rats by cecal ligation and puncture(CLP),and Huaxian CapsuleⅡ(10 ml/kg)was administered after the operation.Serum levels of IL-1β,TNF-αand LDH,as well as myocardial enzymes of CK-MB were determined after the treatment.The indicators of oxidative stress in myocardium tissue,MDA,GPx and SOD were measured as well after the treatment.Results:The serum levels of IL-1β,TNF-α,LDH and CK-MB could be reduced by the intervention of Huaxian CapsuleⅡ.Compared to the CLP group,the treatment group could significantly inhibit the oxidative stress of myocardial tissue and increase the antioxidant level(P<0.01).Conclusion:Huaxian CapsuleⅡcan reduce the oxidative stress in the rats with sepsis-induced myocardium injury.
