摘 要:目的:建立同时测定十全大补丸中阿魏酸、芍药苷、毛蕊花糖苷、桂皮醛、甘草苷、甘草酸铵、党参炔苷和黄芪甲苷的含量测定方法。方法:采用HPLC法,色谱柱为Agilent HC-C18(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速:1.0 ml·min-1;检测波长:230 nm(芍药苷)、237 nm(甘草苷、甘草酸铵)、330 nm(毛蕊花糖苷、阿魏酸)、290 nm(桂皮醛)、368 nm (党参炔苷、黄芪甲苷);柱温:30℃,进样量:20μl。结果:阿魏酸、芍药苷、毛蕊花糖苷、桂皮醛、甘草苷、甘草酸铵、党参炔苷和黄芪甲苷检测质量浓度的线性范围分别为5.00~49.50μg·ml-1、18.00~180.20μg·ml-1、3.30~32.50μg·ml-1、5.00~50.30μg·ml-1、6.00~60.10μg·ml-1、3.10~30.70μg·ml-1、27.30~273.30μg·ml-1、10.30~102.80μg·ml-1(r为0.999 3~0.999 9);平均加样回收率分别为99.2%,101.9%,98.2%,101.1%,98.4%,98.8%,97.2%,100.1%(RSD<2.0%,n=6)。结论:该方法准确、可靠,重复性好,可用于十全大补丸的质量评价。Objective: To develop a method for the simultaneous determination of ferulic acid,paeoniflorin,verbascoside,cinnamaldehyde,liquiritin,ammonium,lobetyolin and astragaloside in Shiquan Dabu pills by HPLC. Methods: The analysis was carried out on an Agilent HC-C18 column(250 mm×4.6 mm,5 μm) with the mobile phase consisting of acetonitrile-0.1% phosphoric acid in a gradient elution mode at the flow rate of 1.0 ml · min-1. The detection wavelength was set at 230 nm for paeoniflorin,237 nm for liquiritin and ammonium,330 nm for verbascoside and ferulic acid,290 nm for cinnamaldehyde,368 nm for lobetyolin and astragaloside. The column temperature was set at 30 ℃ and the injection volume was 20 μl. Results: The linear range was 5.00-49.50μg·ml-1,18.00-180.20μg·ml-1,3.30-32.50μg·ml-1,5.00-50.30μg·ml-1,6.00-60.10 μg·ml-1,3.10-30.70μg·ml-1,27.30-273.30μg·ml-1 and 10.30-102.80 μg·ml-1 for ferulic acid,paeoniflorin,verbascoside,cinnamaldehyde,liquiritin,ammonium,lobetyolin and astragaloside,respectively(r = 0.999 3-0.999 9). The average recovery was 99.2%,101.9%,98.2%,101.1%,98.4%,98.8%,97.2% and 100.1%(RSD<2.0%,n = 6),respectively. Conclusion: The method is simple,accurate,sensitive and reliable in the quality evaluation of Shiquan Dabu pills.
