摘 要:目的探究木槿花(flos hibisci)多糖的降血糖作用机制。方法采用高脂饲养结合ip低剂量STZ制备Ⅱ型糖尿病模型,随机分为空白组、模型组,二甲双胍组,木槿花多糖(flos hibisci polysaccharide,FHP)低、中、高剂量组,在连续口服给药16 w后进行空腹血糖,口服葡萄糖糖耐量(orat glucose tolerance test,OGTT)测定、血清和肝组织中相关生化指标检测。结果FHP能降低Ⅱ型糖尿病小鼠的血糖、食物消耗量和水摄取量,增加Ⅱ型糖尿病小鼠体重;降低Ⅱ型糖尿病小鼠血清中的TG、TC和LDL水平,增加HDL水平,改善Ⅱ型糖尿病血脂症状;增加肝脏和血清中的SOD、CAT和GPX水平,降低MDA和ROS水平,改善氧化应激水平;能激活肝脏中的PI3K/AKT/GSK3β信号通路,改善肝糖原代谢。结论木槿花多糖通过改善血脂代谢、氧化应激水平、调控PI3K/AKT/GSK3β信号通路,改善肝糖原代谢发挥降糖作用。Objective To investigate the hypoglycemic mechanism of Flos hibisci polysaccharide.Methods The mouse models of type 2 diabetes mellitus was established by high-fat feeding combined with ip low-dose STZ and randomly divided into blank group,model group,metformin group,and low,medium and high dose groups of flos hibisci polysaccharide(FHP).The fasting blood glucose,oral glucose tolerance(OGTT),and related biochemical indicators in serum and liver tissues were measured after continuous oral administration for 16 weeks.Results FHP reduced the blood glucose,food consumption and water intake of type 2 diabetic mice and increased their body weight.It also reduced their levels of TG,TC and LDL in serum,increased their HDL levels,and lowered the blood fat.FHP increased the SOD,CAT and GPX levels in liver and serum and reduced the MDA and ROS levels,improving the level of oxidative stress.In addition,it activated PI3K/AKT/GSK3βsignaling pathway in liver and improved hepatic glycogen metabolism.Conclusion Flos hibisci polysaccharide plays a hypoglycemic role by improving blood lipid metabolism,oxidative stress level and hepatic glycogen metabolism and regulating PI3K/AKT/GSK3βsignaling pathway.
