摘 要:以甲醇、95%乙醇、80%丙酮、丙酮-乙醇混合液(体积比1∶1)、二甲基亚砜为提取液,分别提取金线莲叶片叶绿素,用比色法测定叶绿素含量,旨在筛选叶绿素测定适宜的浸提液。结果表明:叶绿素a测定值由高到低依次为80%丙酮(3.700)、二甲基亚砜(3.686)、丙酮-乙醇(3.585)、甲醇(3.448)、95%乙醇(3.436),其中80%丙酮、二甲基亚砜测定值极显著高于甲醇、95%乙醇测定值,丙酮-乙醇测定值极显著高于95%乙醇测定值;叶绿素b测定值由高到低依次是80%丙酮(1.505)、丙酮-乙醇(1.471)、95%乙醇(1.418)、甲醇(1.365)、二甲基亚砜(1.280),其中80%丙酮、丙酮-乙醇、95%乙醇、甲醇测定值极显著或显著高于二甲基亚砜测定值;总叶绿素测定值由高到低分别是80%丙酮(5.206)、丙酮-乙醇(5.057)、二甲基亚砜(4.966)、95%乙醇(4.854)、甲醇(4.813),其中80%丙酮测定值显著高于二甲基亚砜测定值,极显著高于95%乙醇、甲醇提取测定值,丙酮-乙醇测定值显著高于95%乙醇测定值,极显著高于甲醇测定值;叶绿素a/b值测定结果,二甲基亚砜测定值2.880,极显著高于甲醇、80%丙酮、丙酮-乙醇和95%乙醇测定值2.527、2.459、2.437和2.421。综合比较分析,金线莲叶绿素含量测定提取液以80%丙酮或丙酮-乙醇混合液为宜。Extracts,methanol,95%ethanol,80%acetone,acetone ethanol mixture(1:1)and dimethyl sulfoxide(DMSO)were used to extract chlorophyll from Anoectophora roxburghii leaves,and the content of chlorophyll was determined by colorimetry in order to screen the suitable extract for extraction of chlorophyll.The extraction of chlorophyll a from the A.roxburghii leaves showed that the contents of chlorophyll a varied in the order of 80%acetone(3.700)>DMSO(3.686)>acetone ethanol(3.585)>methanol(3.448)>95%ethanol(3.436).The chlorophyll a contents extracted by 80%acetone and DMSO are highly significantly higher than those by methanol and 95%ethanol,and the chlorophyll a content extracted by acetone ethanol mixture is highly significantly higher than that by 95%ethanol.The extraction of chlorophyll b from the A.roxburghii leaves showed that the chlorophyll b contents varied in the order of 80%acetone(1.505)>acetone ethanol(1.471)>95%ethanol(1.418)>methanol(1.365)>DMSO(1.280).The chlorophyll b contents extracted by 80%acetone,acetone ethanol,95%ethanol,and methanol are highly significantly higher or significantly higher than that by DMSO.Extraction of total chlorophyll showed that the total chlorophyll contents varied in the order of 80%acetone(5.206)>acetone ethanol(5.057)>DMSO(4.966)>95%ethanol(4.854)>methanol 4.813.The total chlorophyll content extracted by 80%acetone is significantly higher than that by DMSO,and highly significantly higher than those by 95%ethanol and methanol.The total chlorophyll content extracted by acetone ethanol is significantly higher than that by 95%ethanol and highly significantly higher than that by methanol.The chlorophyll a/b ratio extracted by DMSO(2.880)is significantly higher than those by methanol,80%acetone,acetone ethanol mixture and 95%ethanol(2.527,2.459,2.437 and 2.421,respectively).Comprehensive comparative analysis showed that 80%acetone or acetone ethanol mixture is the best extraction solution for extraction of chlorophyll content of A.roxburghii.
