摘　　要：目的:观察围着床期小鼠子宫内膜同源盒基因A10(HOXA10)、整合素αvβ3和白血病抑制因子(LIF)的表达,探讨控制性超排卵(COH)对小鼠子宫内膜容受性的影响。方法:昆明纯种性成熟小鼠经COH模型制备、胚泡移植后,随机分为自然囊胚+自然假孕组、COH囊胚+自然假孕组、自然囊胚+COH假孕组、COH囊胚+COH假孕组。取受孕第6天小鼠,观察各组妊娠率及平均着床位点数,采用Western blot和RT-PCR技术检测受孕第5天小鼠子宫内膜HOXA10、整合素αvβ3和LIF蛋白及mRNA的表达。结果:COH假孕鼠接受囊胚移植后妊娠率、平均着床位点数较自然假孕鼠低;子宫内膜HOXA10、整合素αvβ3和LIF表达显著性低于自然假孕鼠,差异有统计学意义(P<0.01)。结论:超排卵药物能下调种植窗期子宫内膜HOXA10、αvβ3和LIF的表达,降低子宫内膜容受性,导致临床妊娠率低下。Objective:To observe the expression of endometrial homologous box gene A10(HOXA10),integrinαvβ3 and leukemia suppressor factor(LIF)in mice during peri-implantation period,and to investigate the effect of controlled superovulation(COH)on endometrial receptivity in mice.Methods:After establishement of COH model and blastocyst transplantation,Kunming pure-bred mature mice were randomly divided into the natural blastocyst+natural pseudocyst group,the COH blastocyst+natural pseudocyst group,the natural blastocyst+COH pseudocyst group,and the COH blastocyst+COH pseudocyst group.The pregnancy rate and average number of beds in each group were observed on day 6 of conception.Western blot and RT-PCR were used to detect the protein and mRNA expression of HOXA10,integrinαvβ3,LIF in the endometria of mice on day 5 of conception.Results:The pregnancy rate and average number of beds in the COH group were lower than those in the natural receptor group.The expressions of HOXA10,integrinαvβ3 and LIF in endometrium were significantly lower than these of the natural receptor mice(P<0.01).Conclusion:Superovulation drugs could down-regulate the expression of HOXA10,integrinαvβ3,and LIF in endometrium during the implantation window,reduce endometrial receptivity,and lead to low clinical pregnancy rate.
