摘  要:目的探讨丹参酮ⅡA(TA)和丹酚酸(SA)B联合用药对成纤维细胞转化的影响。方法Transwell小室法检测TA和SAB对血管紧张素(Ang)Ⅱ诱导的细胞迁移的影响;Western印迹检测成纤维细胞α-平滑肌肌动蛋白(SMA)的表达;流式细胞术检测活性氧(ROS)水平的变化。结果TA和SAB组合(T8S5)对成纤维细胞的迁移能力有显著性抑制作用(P<0.01);T8S5对AngⅡ诱导的α-SMA的表达有显著抑制作用(P<0.01);T8S5能减少成纤维细胞内ROS的表达(P<0.01)。结论SAB和TA配伍组合能抑制AngⅡ诱导的大鼠血管外膜成纤维细胞的迁移和转化。Objective To explore the effects of tanshinoneⅡA(TA)and salvianolic acid(SA)B in combination on the differentiation of rat vascular adventitial fibroblasts.Methods The effect of TA and SAB in combination on the migration of adventitial fibroblast induced by Angiotensinogen(Ang)Ⅱwere evaluated by Transwell,and the effect of TA and SAB in combination on the expression ofα-smooth muscle actin(SMA)was detected by Western blot.The reactive oxygen species(ROS)level was detected by flow cytometry.Results The results of Transwell showed that TA and SAB in combination(T8S5)inhibited the migration of fibroblasts(P<0.01).The results of Western blot showed that T8S5 significantly inhibited the expression ofα-SMA induced by AngⅡ(P<0.01).Flow cytometry showed that T8S5 decreased the expression of ROS(P<0.01).Conclusions TA and SAB in combination could inhibit AngⅡ-induced migration and transformation of rat adventitial fibroblasts.
