摘  要:目的探讨神经调节蛋白1/转化生长因子β1(NRG-1/TGF-β1)对血管平滑肌细胞骨架的作用。方法取小鼠血管组织,采用免疫荧光法对NRG-1干预后细胞收缩功能变化进行分析,采用鬼笔环肽、免疫荧光共定位分析NRG-1/TGF-β1表达,采用GST融合蛋白沉降技术检测分析NRG-1-ICD与α-肌动蛋白(α-SMA)的共位性。结果 TGF-β1在人主动脉血管平滑肌细胞中可上调NRG-1表达,经免疫荧光染色可发现平滑肌细胞标志蛋白α-SMA与NRG-1存在共位性;经RT-PCR及Western blot检测分析发现,在大鼠平滑肌细胞、人平滑肌细胞以及人内皮细胞中均存在NRG-1的mRNA表达。结论在转录和翻译水平上TGF-β1可上调NRG-1的表达;在细胞收缩中,TGF-β1对于α-SMA与NRG-1-ICD之间的表达可以起到促进作用。Objective To investigate the effect of neuregulin 1/transforming growth factorβ1(NRG-1/TGF-β1)on the cytoskeleton of vascular smooth muscle.Methods Blood vessel tissues of mice were taken,and the changes of cell contractile function after NRG-1 intervention were analyzed by immunofluorescence method,and the expression of NRG-1/TGF-β1 was analyzed by phalloidin and immunofluorescence co-localization,and the GST fusion protein was used for sedimentation.Technical detection and analysis of the co-location of NRG-1-ICD andα-actin(α-SMA)were performed.Results TGF-β1 could up-regulate the expression of NRG-1 in human aortic vascular smooth muscle cells.Immunofluorescence staining showed that the smooth muscle cell marker proteinα-SMA and NRG-1 were co-located;RT-PCR and Western blot found that NRG-1 mRNA expression existed in rat smooth muscle cells,human smooth muscle cells and human endothelial cells.Conclusion TGF-β1 can up-regulate the expression of NRG-1 at the transcription and translation levels;in cell contraction,TGF-β1 can promote the expression ofα-SMA and NRG-1-ICD.
