摘 要:目的:探讨蟛蜞菊内酯联合蚯蚓纤溶酶对高同型半胱氨酸血症(Hyperhom Ocysteinemia,Hhcy)大鼠血管的保护作用。方法:将SPF级雄性SD大鼠60只,随机分为6组:正常对照组、模型对照组、阳性药物组、蟛蜞菊内酯(Wedelolactone,Wed)药物组、蚯蚓纤溶酶(Earthworm Fibrinolyticl Enzyme,EFE)药物组和Wed+EFE药物组。正常组喂饲普通饲料,其余各组喂饲高L-蛋氨酸饲料(2%L-蛋氨酸+普通饲料)。各组均采用每日灌胃给药,正常组和模型组给予生理盐水,阳性药物组给予联合用药,叶酸、维生素B12(VitB12)和维生素B6(VitB6)用量分别为0.49 mg/kg、2.47 mg/kg、0.49 mg/kg,其余各组分别以Wed(100 mg/kg)、EFE(1000μku/kg)、Wed+EFE灌胃,8周后比色法测定同型半胱氨酸(Homocysteine,Hcy)、白介素1α(Interleukin-1α,IL-1α)和白介素1β(Interleukin-1β,IL-1β)、血清胆固醇(Total Cholesterol,TC),三酰甘油(Triglyceride,TG)、低密度脂蛋白(Low Density Lipoprotein Cholesterol,LDL-C)、高密度脂蛋白(High-Density Lipoprotein Cholesterol,HDL-C)水平。结果:蟛蜞菊内酯和蚯蚓纤溶酶单独作用时,对降低大鼠Hcy、IL-1α、IL-1β,控制血浆中TC、TG、LDL-C水平升高,均显示出一定作用,Wed+EFE联合作用可有效控制血浆Hcy、炎症介质IL-1α和IL-1β的升高,对降低TC、TG和LDL-C也有很好的作用,且能明显升高血浆HDL-C水平,这一点优于阳性药物对照组。结论:蟛蜞菊内酯和蚯蚓纤溶酶的联合应用能够降低血浆中Hcy、IL-1α和IL-1β水平,降低TC、TG和LDL-C水平,并有效提升血浆HDL-C水平,这将有利于临床控制(Atherosclerosis,AS)的发生和发展。Objective:To investigate the protective effects of wedelolactone combined with earthworm fibrinolysin on the vessels of rats with hyperhomocysteinemia.Methods:Sixty SPF male SD rats were randomly divided into 6 groups:Normal Control Group,Model Control Group,positive drug group,Wed group,EFE Group and Wed+EFE group.The normal group was fed with normal diet and the other groups were fed with high L-methionine Diet(2%L-methionine+normal diet).The drags were administrated by intragastric administration once a day.The normal group and the model group were given saline solution,and the positive group was given combined medication.The dosage of folic acid,VitB12 and VitB6 were 0.49,2.47 and 0.49 mg/kg respectively,the other groups were given with Wed(100 mg/kg),EFE(1000μku/kg),Wed+EFE.After 8 weeks,the levels of Homocysteine,IL-1-(IL-1)and IL-1-(IL-1),serum cholesterol(TC),triglyceride(TG),Low-density Lipoprotein(LDL-C)and high-density Lipoprotein(HDL-C)were measured by colorimetry.Results:When wedelolactone and earthworm fibrinolysin act alone,they can reduce the levels of Hcy,IL-1αand IL-1β,and control the increase of TC,TG and LDL-C in plasma of rats.The combined action of Wed+Efe could effectively control the increase of Hcy,IL-1 and IL-1 in plasma,decrease TC,TG and LDL-C,and increase HDL-C in plasma significantly,which was superior to the positive drug control group.Conclusion:The combination of wedelolactone and earthworm fibrinolysin can decrease the levels of Hcy,IL-1 and IL-1 in plasma,decrease the levels of TC,TG and LDL-C,and increase the level of HDL-C in plasma.This will be conducive to clinical control of the occurrence and development of AS.
