摘 要:目的:探讨葡萄籽原花青素对脑缺血再灌注损伤的保护作用。方法:将72只大鼠随机分为假手术组、模型组、原花青素干预组,8周后假手术组行麻醉及血管分离术,模型组和干预组构造线栓法局灶性大鼠大脑中动脉梗塞模型。大鼠缺血2小时再灌注24 h后,分别测定神经功能缺损评分、大鼠脑梗死体积、脑含水量及脑组织中CD40配体(CD40 Ligand,CD40L)及基质金属蛋白酶-9(Matrix Metalloproteinase-9,MMP-9)表达水平。结果:假手术组脑组织中CD40L及MMP-9均未见表达,而模型组及干预组均有表达,且模型组中CD40L及MMP-9的表达水平均高于干预组,差异有统计学意义(P<0.01)。结论:葡萄籽原花青素预处理能改善大鼠脑缺血再灌注损伤后的神能功能缺损,减轻脑水肿及脑梗死体积,能通过降低脑组织中CD40L及MMP-9的表达水平,从而保护脑缺血再灌注损伤。Objective:To study the neuroprotective mechanism of grape seed proanthocyanidin extracts against the injury induced by cerebral ischemia-reperfusion.Methods:72 rats were randomly divided into the sham-operation group(Sham group),the ischemiareperfusion group(IR group),the grape seed procyanidins treatment group(GSPE group/intervention group),after eight weeks feeding,sham-operation group received line anesthesia and vascular separation technique,the model group and the intervention group constructed line bolt method foci of rat middle cerebral artery infarction model,after rats ischemia 2 hours reperfusion 24 hours,measured the cerebral infarction volume,cerebral water content,nerve function defect scale and the expression of CD40L,MMP-9 in brain tissue after ischemia reperfusion injury.Results:Sham group showed no CD40L and MMP-9 expression in brain tissue,but CD40L and MMP-9 expressed in the model group and the intervention group,and the expression level of CD40L and MMP-9 of the model group was higher than that of the intervention group,the difference was of statistical significance(P<0.01).Conclusion:Grape seed procyanidins pretreatment can improve the cerebral infarction volume,cerebral water content and nerve function defect scale in rats with high fat diet after ischemia reperfusion injury,and through reducing the expression level of CD40L and MMP-9 in brain tissue after ischemia reperfusion injury,it protects cerebral ischemia reperfusion injury.
