不同糖代谢人群中Txnip水平及其与炎症/氧化应激的相关性研究

摘　　要：目的观察不同糖代谢人群硫氧还蛋白相互作用蛋白(Txnip)的水平,并研究其与炎症/氧化应激的相关性。方法选择87例健康体检者为研究对象,根据糖代谢水平不同,分为3组:糖尿病组(DM组)27例;糖尿病前期组(PD组)30例;正常糖耐量组(NGT组)30例。收集研究对象的一般资料,检测空腹血糖、空腹胰岛素、OGTT 2 h血糖(2 h-PBG),并计算胰岛素抵抗指数(HOMA-IR)和胰岛β细胞敏感指数(HOMA-β),测定血液中Txnip、IL-1β、IL-18、MDA、SOD水平。观察上述指标在不同糖代谢人群中的变化规律,并分析上述指标间的相关性。结果DM组体重及体质指数均高于NGT组(P<0.05)。3组间空腹血糖、2 h-PBG、空腹胰岛素、HOMA-IR、Txnip、IL-1β、IL-18及MDA比较,DM组>PD组>NGT组(P<0.05)。3组间HOMA-β、硫氧还蛋白(Trx)、SOD比较,NGT组>PD组>DM组(P<0.05)。Txnip与IL-1β(r=0.761,P<0.05)、IL-18(r=0.796,P<0.05)、MDA(r=0.628,P<0.05)呈正相关性,与Trx(r=-0.770,P<0.05)、SOD(r=-0.661,P<0.05)呈负相关性。Trx与SOD(r=0.395,P<0.05)呈正相关性,与L-1β(r=-0.667,P<0.05)、IL-18(r=-0.715,P<0.05)、MDA(r=-0.300,P<0.05)呈负相关性。结论Txnip在糖尿病前期人群中即表达增加,Txnip与机体氧化应激及炎症损伤密切相关。Objective To observe the level of thioredoxin interacting protein(Txnip)in people with different glycometabolism and to study the correlation with inflammation/oxidative stress.Methods 87 healthy people were randomly divided into three groups according to the different levels of glucose metabolism:diabetes group(DM,n=27),prediabetes group(PD,n=30),normal glucose tolerance group(NGT,n=30).The general data of the subjects were collected and the fasting blood glucose,fasting insulin and OGTT 2h blood glucose(2h-PBG)were detected,and HOMA-IR and HOMA-βcell function(HOMA-β)were calculated,the levels of Txnip,IL-1β,IL-18,MDA and SOD in blood were measured.The correlation between the above indexes were analyzed.Results The body weight and body mass index of DM group were higher than those of NGT group(P<0.05).Fasting blood glucose,2 h-PBG,fasting insulin,HOMA-IR,Txnip,IL-1β,IL-18 and MDA were compared among the three groups,DM>PD>NGT(P<0.05).Compared with HOMA-β,TRX and SOD among the three groups,NGT>PD>DM(P<0.05).Txnip was positively correlated with IL-1β(r=0.761,P<0.05),IL-18(r=0.796,P<0.05),MDA(r=0.628,P<0.05),and negatively correlated with TRX(r=-0.770,P<0.05),SOD(r=-0.661,P<0.05).TRX was positively correlated with SOD(r=0.395,P<0.05),and negatively correlated with L-1β(r=-0.667,P<0.05),IL-18(r=-0.715,P<0.05)and MDA(r=-0.300,P<0.05).Conclusion The expression of Txnip is increased in prediabetes.Txnip is closely related to oxidative stress and inflammatory injury in diabetic patients..
