摘  要:目的观察超声微泡爆破辅助骨髓间充质干细胞(BMSC)移植治疗大鼠慢性肾病(CKD)。方法40只SD大鼠随机分为4组各10只。BMSC+微泡组、BMSC组、CKD组均制备CKD模型,制模后BMSC+微泡组大鼠经肾动脉注射0.5 mL的BMSC(2×10^(6)/mL),随之注入0.5 mL声诺维造影剂微泡(2×10^(8)个/毫升),注射完毕后立即启动西门子S3000超声诊断仪对两侧肾脏每隔10 s分别爆破1次,共3次;BMSC组大鼠经肾动脉注射0.5 mL的BMSC(2×10^(6)/mL)悬液;对照组及CKD组大鼠经肾动脉输注等量0.9%氯化钠溶液。采用自动生化分析仪检测各组大鼠治疗前和治疗2周后血清BUN、血清Cr、24 h尿蛋白,HE染色及Masson三色染色评估治疗2周后肾脏组织病理变化。结果与同组治疗前比较,BMSC+微泡组大鼠血清Cr、血清BUN、24 h尿蛋白水平降低,BMSC组大鼠血清Cr、24 h尿蛋白水平降低,CKD组大鼠治疗后血清Cr、24 h尿蛋白水平升高(P均<0.05)。与对照组比较,BMSC组大鼠治疗后血清BUN及CKD组治疗后血清Cr、血清BUN、24 h尿蛋白水平升高(P均<0.05)。与CKD组比较,BMSC+微泡组和BMSC组大鼠血清Cr、血清BUN、24 h尿蛋白水平均降低(P均<0.05)。与BMSC组比较,BMSC+微泡组大鼠血清Cr、24 h尿蛋白水平降低(P均<0.05)。与CKD组比较,BMSC组HE染色及Masson三色染色显示CKD样病理有所改善,BMSC+微泡组CKD样病理改善更为明显。结论超声微泡爆破联合BMSC移植治疗大鼠CKD效果较好,其可减轻大鼠CKD肾脏损伤程度,改善肾脏功能。Objective To observe the effect of ultrasound microbubble blasting combined with bone marrow mesenchymal stem cells(BMSC)transplanted to treat chronic kidney disease(CKD)in rats.Methods Forty SD rats were randomly divided into 4 groups with 10 rats in each group.CKD models were prepared in the BMSC+microbubble group,BMSC group,and CKD group.After making the model,rats in the BMSC+microbubble group were injected with 0.5 mL of BMSC(2×10^(6)/mL)through the renal artery,followed by 0.5 mL of sonovir contrast agent Microbubbles(2×10^(8) pcs/mL).Immediately after the injection,the Siemens S3000 ultrasonic diagnostic apparatus was activated to blast the kidneys on both sides once for every 10 s,a total of 3 times;rats in the BMSC group were injected with 0.5 mL of BMSC(2×10^(6)/mL)suspension;rats in the control group and CKD group were infused with an equal amount of 0.9%sodium chloride solution through the renal artery.The automatic biochemical analyzer was used to detect serum BUN,serum Cr,and 24 hour urine protein of rats in each group before treatment and 2 weeks after treatment.HE staining and Masson trichrome staining were used to evaluate the pathological changes of kidney tissues after 2 weeks of treatment.Results Compared with rats in the same group before treatment,the levels of serum Cr,serum BUN,and 24 hour urine protein in rats in the BMSC+microvesicle group decreased,while the levels of serum Cr and 24 hour urine protein in rats decreased in the BMSC group.Levels of serum Cr and 24 hour urine protein in rats in CKD group were increased after treatment(all P<0.05).Compared with the control group,the levels of serum BUN in the BMSC group,and the levels of serum Cr,serum BUN and24 hour urine protein in the CKD group increased after treatment(all P<0.05).Compared with the CKD group,the serum Cr,serum BUN,and 24 hour urine protein levels in the BMSC+microvesicle group and the BMSC group decreased(all P<0.05).Compared with the BMSC group,the levels of serum Cr and 24 hour urine protein in the BMSC+microbubbl
