摘 要:目的观察宁动颗粒对抽动-秽语综合征(TS)模型大鼠抽动刻板和认知行为的影响。方法24只Wistar TS大鼠随机分为阿朴吗啡组(CTR,n=8)、氟哌啶醇组(Hal,n=8)和宁动颗粒组(NDG,n=8),均腹腔注射阿朴吗啡,诱导出抽动症状,随机抽取两组造模同时分别给予氟哌啶醇和宁动颗粒。3组模型分别进行抽动行为、开场试验和Morris水迷宫实验监测,观察药物干预后对行为学的影响。结果药物干预第5周后,Hal组和NDG组与CTR组比较刻板活动明显减弱(P<0.05),且第8周后差异最大,Hal组和NDG组比较差异无统计学意义。干预后,NDG组大鼠在中央停留时间显著增加,穿越格子数、直立次数和修饰次数显著减少(P<0.05),而Hal组均无明显变化。3组大鼠经多次练习,找到平台的时间均减少,与第1天比较差异有统计学意义(P<0.05),NDG组找到平台的时间较Hal组和CTR组改善更明显。结论宁动颗粒能有效抑制TS模型大鼠抽动等刻板行为,并改善TS模型大鼠多动及学习记忆能力。Objective To observe the effect of Ningdong granule on stereotype and cognitive behavior of tourette’s syndrome(TS)model rats.Methods 24 Wistar TS rats were randomly divided into apomorphine group(CTR,n=8),haloperidol group(HAL,n=8)and Ningdong granule group(NDG,n=8).All rats were intraperitoneally injected with apomorphine to induce tic symptoms.Two groups were randomly selected for modeling and were given haloperidol and Ningdong granules respectively.The twitch behavior monitoring,opening test and Morris water maze test were carried out to observe the effect on behavior among three groups.Results After 5 weeks of drug intervention,compared with CTR group,the stereotype activity of Hal group and NDG group was significantly reduced(P<0.05),and the difference was the largest after 8 weeks.There was no significant difference between Hal group and NDG group.After the intervention,the time of staying in the center of NDG group was significantly increased,the number of crossing the grid,the number of standing and the number of modification were significantly decreased(P<0.05),but there was no significant change in Hal group.After repeated practice,the time to find the platform in the three groups was reduced,and the difference was statistically significant compared with the first day(P<0.05).The time to find the platform in the NDG group was better than that in the Hal group and CTR group.Conclusion Ningdong granule can effectively inhibit the tic and other stereotyped behaviors of TS model rats,and improve the hyperactivity and learning and memory ability of TS model rats.
