摘  要:本试验旨在探讨高、低淀粉饲粮对山羊小肠黏膜结构、肠道激素浓度、甜味受体及其通路元件基因表达的影响。试验选择体况相似的雄性湘东成年黑山羊20只,随机分为2组,每组10只,分别饲喂高淀粉(50.4%,HS组)和低淀粉饲粮(13.1%,LS组)。试验期38 d。结果表明:1)与LS组相比,HS组空肠和回肠的隐窝深度增加(P>0.05),十二指肠和空肠的绒毛高度降低(P>0.05);回肠黏膜中胆囊收缩素(CCK)(P>0.05)和胰高血糖素样肽-2(GLP⁃2)(P>0.05)及回肠内容物中胰高血糖素样肽-1(GLP⁃1)(P<0.05)和酪酪肽(Pyy)(P>0.05)的浓度均增加,但回肠黏膜和回肠内容物中胃泌素抑制肽(GIP)的浓度降低(P>0.05)。2)2组间Ⅰ型味觉受体3(T1R3)、α-味移导素(α⁃gustducin)、瞬时受体电位离子通道蛋白M型5(TRPM5)、葡萄糖转运蛋白2(GLUT2)和钠/葡萄糖共转运载体1(SGLT1)在十二指肠、空肠和回肠中的基因表达量无显著差异(P>0.05),但HS组空肠α⁃gustducin基因表达量显著低于LS组(P<0.05)。3)相关性分析显示,回肠内容物葡萄糖浓度与回肠α⁃gustducin、TRPM5和SGLT1的基因表达量之间呈显著正相关(0.05<r<0.30,P<0.05),而与空肠甜味受体关键元件T1R3与SGLT1的基因表达量之间的相关系数为0.93(P<0.01),与空肠TRPM5基因表达量之间的相关系数为0.66(P<0.01),与空肠GLUT2基因表达量之间的相关系数为0.48(P<0.01)。综上所述,相比粗蛋白质和粗纤维,淀粉是甜味信号通路元件以及葡萄糖转运载体的主要影响因素,但高淀粉饲粮对发育成熟的山羊肠道黏膜结构及功能影响较小。This experiment was to explore the impact of high and low starch intake on the small intestinal mu⁃cosal structure,gastrointestinal(GI)hormone concentrations,sweet taste receptors(STR)and the key ele⁃ments of STR signaling pathway in goats.Twenty mature male Liuyang black goats were randomly allocated to 2 groups and 10 goats per group.HS group fed a high starch content(50.4%),and the LS group fed a low starch content(13.1%).The experiment lasted for 38 d.The results showed as follows:1)compared with LS group,the crypt depth of jejunum and ileum in HS group increased(P>0.05),and the villus height of duode⁃num and jejunum decreased(P>0.05);the concentrations of cholecystokinin(CCK)(P>0.05),glucagon like peptide⁃2(GLP⁃2)(P>0.05)in ileal mucosa,and glucagon like peptide⁃1(GLP⁃1)(P<0.05)and pep⁃tide YY(Pyy)(P>0.05)in ileal chyme increased,but gastrin inhibitory peptide(GIP)in ileal mucosa and chyme decreased(P>0.05).2)There was no significant difference in the gene expression of 3 typeⅠtaste receptors(T1R3),α⁃gustducin,transient receptor potential ion channel protein M type 5(TRPM5),glucose transporter 2(GLUT2)and sodium/glucose co⁃transporter 1(SGLT1)in duodenum,jejunum and ileum(P>0.05).In jejunum,gene expression ofα⁃gustducin in LS group was significantly higher than that in HS group(P<0.05).3)Correlation analysis showed that ileal glucose concentration was positively correlated with gene expressions ofα⁃gustducin,TRPM5 and SGLT1 of ileal(0.05<r<0.30,P<0.05).In addition,the gene ex⁃pression of T1R3 in jejunum was high related(P<0.01)to that of SGLT1,TRPM5 and GLUT2 in jejunum with the correlation efficiency as 0.93(P<0.01),0.66(P<0.01)and 0.48(P<0.01),respectively.In con⁃clusion,compared with the crude protein and fiber in diets,starch is the major factor on the gene expressions of sweet taste receptors and glucose transporters,but with a little effect on the intestinal mucosal structure.
