摘 要:目的:探讨发酵虫草菌粉(Cs-4)对糖尿病肾脏疾病大鼠肾功能的保护作用及初步机制。方法:采用高糖高脂饲料喂养联合腹腔注射链脲佐菌素(40 mg·kg^(-1))的方法复制糖尿病肾脏疾病大鼠模型。将所有模型大鼠随机分为模型组、Cs-4低(300 mg·kg^(-1))、中(600 mg·kg^(-1))、高剂量组(1200 mg·kg^(-1))及阳性药组(厄贝沙坦,mg·kg^(-1)),同时设正常组(给予普通饲料)和高糖高脂组(给予高糖高脂饲料),并均给予腹腔注射相同剂量的柠檬酸缓冲液。各组大鼠每天灌胃(ig)给药1次,连续8周。实验结束收集24 h内尿液,测定大鼠空腹血糖。收集大鼠血清、肾脏,以试剂盒法检测大鼠血清尿素氮、血肌酐、超氧化物歧化酶、丙二醛、总胆固醇、甘油三酯、低密度脂蛋白、高密度脂蛋白、白介素6、白介素8、肿瘤坏死因子α、超敏c反应蛋白、游离脂肪酸、糖化血清蛋白、单核细胞趋化蛋白1、转化生长因子β1及尿液尿肌酐、尿蛋白、尿微量白蛋白含量,以苏木精-伊红染色观察大鼠肾组织病理形态学变化。结果:与模型组相比,Cs-4各剂量组在不同程度上可降低肾重、肾重/体重、空腹血糖、尿素氮、血肌酐、尿蛋白、尿微量白蛋白、总胆固醇、低密度脂蛋白、丙二醛、白介素6、白介素8、肿瘤坏死因子α、超敏c反应蛋白、游离脂肪酸、糖化血清蛋白、单核细胞趋化蛋白1、转化生长因子β1水平,增加体重,提高尿肌酐、内生肌酐清除率、高密度脂蛋白、超氧化物歧化酶水平(P<0.05,P<0.01),且Cs-4高剂量组作用更显著,在不同程度上能改善肾组织形态损伤。结论:Cs-4对糖尿病肾脏疾病大鼠肾功能损害具有一定的保护作用,其机制可能与降低血糖、调节血脂、改善氧化应激及炎症水平、抑制肾脏纤维化程度有关。Objective:To explore the protective effect of fermented Cordyceps Sinensis powder(Cs-4)on renal function of diabetic kidney disease rats and preliminarily investigate the mechanism.Methods:The rat model of diabetic kidney disease was replicated by high-sugar and high-fat diet feeding combined with intraperitoneal injection of streptozotocin(40 mg·kg^(-1)).The model rats were randomly divided into model group,low(300 mg·kg^(-1)),medium(600 mg·kg^(-1))and high dose(1200 mg·kg^(-1))of Cs-4 groups and positive drug group(irbesartan,15 mg·kg^(-1)).At the same time,the normal group(receiving ordinary diet)and the high-sugar and high-fat groups(receiving high-sugar and high-fat diet)were set up,and they were given intraperitoneal injection of the same dose of citrate buffer.Rats in each group were administered by intragastric(ig)administration once a day for 8 consecutive weeks.At the end of the experiment,urine was collected within 24 hours,and the fasting plasma glucose level was measured.Serum and kidney were collected,and kit method was used to detect blood urea nitrogen,serum creatinine,superoxide dismutase,malondialdehyde,total cholesterol,triglycerides,low density lipoprotein,high density lipoprotein,interleukin 6,interleukin 8,tumor necrosis factorα,high sensitivity c-reactive protein,free fatty acid,glycated serum protein,monocyte chemotactic protein 1,transforming growth factorβ1 in serum and urinary creatinine,urinary protein,microalbuminuria content of rats in each group,and pathological and morphological changes of kidney tissue were observed by hematoxylin-eosin staining.Results:Compared with the model group,all dose groups of Cs-4 reduced kidney weight,kidney weight/body weight ratio,fasting plasma glucose,blood urea nitrogen,serum creatinine,urinary protein,microalbuminuria,total cholesterol,and low density lipid,malondialdehyde,interleukin 6,interleukin 8,tumor necrosis factorα,high-sensitivity c-reactive protein,free fatty acids,glycated serum protein,monocyte chemotactic protein 1,transforming
