摘  要:目的探讨黄精多糖对D-半乳糖(D-Gal)所致衰老小鼠的学习和记忆水平的影响.方法给小鼠皮下注射D-Gal(500 mg/kg),持续8周,建立衰老小鼠模型.将小鼠分为空白组、模型组、阳性药组(吡拉西坦300 mg/kg)和高、中、低剂量的黄精多糖组(600、300、150 mg/kg).空白组注射等体积生理盐水,且模型组和空白组小鼠给予等剂量的蒸馏水灌胃.应用自主活动实验、避暗实验和Morris水迷宫实验方法测定其空间记忆和学习水平,检测小鼠血清中的超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的水平,测定小鼠脑组织中谷氨酸(GLU)、乙酰胆碱(ACh)以及γ-氨基丁酸(GABA)的含量,采用苏木精-伊红(HE)染色的方法观察小鼠海马的CA1区神经元的改变.结果避暗和Morris水迷宫行为学实验表明:与模型组比较,黄精高、中剂量组都可明显改善衰老小鼠学习和记忆的能力(P<0.01或P<0.05),小鼠血清中SOD和GSH-Px的活性增加,MDA含量降低(P<0.05),小鼠脑组织中GLU、ACh含量提高,GABA含量降低(P<0.01或P<0.05).结论黄精多糖能够改善D-Gal衰老小鼠的学习记忆能力.Objective This experiment explored the effect of Polygonatum Sibiricum polysaccharide on learning and memory of aging mice induced by D-galactose(D-Gal).Method Mice were injected D-Gal(500 mg/kg)subcutaneously for 8 weeks.Mice were divided into blank group,model group,positive group(Piracetam 300 mg/kg),high,medium and low doses Polygonatum Sibiricum polysaccharide group(600,300,150 mg/kg),the blank group was injected with an equal volume of normal saline,and mice in the model group and the blank group were given the same doses of distilled water by gavage.The independent activities,dark avoidance and Morris water maze experiments were used to test their dimensional memory and learning ability,and to detect superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione peroxidase(GSH-Px)in mouse serum,the content of glutamate(GLU),acetylcholine(ACh)andγ-aminobutyric acid(GABA)in the brain tissue of mice is tested,and the changes of neurons in the CA1 area of the mouse hippocampus were observed by hematoxylin-eosin(HE)staining.Results The dark avoidance and Morris water maze behavioral experiments showed that:compared with the model group,the high and medium doses of Polygonatum Sibiricum polysaccharide can distinctly improve the learning memory abilities of aging mice(P<0.01 or P<0.05).It can increase SOD and GSH-Px activity in mouse serum,decrease MDA content(P<0.05),increase GLU and ACh content in mouse brain tissue,and reduce GABA content(P<0.01 or P<0.05).It can improve neuronal morphology of D-galactose-induced aging mice.Conclusion The learning memory of aging mice can be better by Polygonatum Sibiricum polysaccharide.
