摘  要:以6个不同甜瓜自交系为试材,测定不同发育时期甜瓜果实中β-胡萝卜素、类胡萝卜素含量和类胡萝卜素代谢相关基因的表达水平,以及葡萄糖、果糖、蔗糖含量和蔗糖代谢相关酶活性的变化。结果表明:甜瓜果实中类胡萝卜素含量与果肉颜色密切相关,随着果实发育时期的延长,自交系156和12B-5-2的果实中β-胡萝卜素和类胡萝卜素含量呈先升高后降低的趋势,B-3-1-1呈升高的趋势,162和159呈降低的趋势。PSY1基因在5个不同肉色的甜瓜果实发育初期表达较高,随着发育时期的延长逐渐下调;PSY2、LCY-b和ZDS基因在自交系162和159中表达趋势与PSY1基因一致,在自交系wm-6、12B-5-2和B3-1-1中表达趋势相反,PSY2、LCY-b和ZDS基因在类胡萝卜素含量最高的wm-6中成熟期表达最强,在类胡萝卜素含量最低的162中表达最弱。果实糖组分和含量与果肉颜色相关性不明显,159果实中葡萄糖含量随着果实发育时期的延长逐渐升高,而成熟期果实中蔗糖含量最低;wm-6、12B-5-2和162果实中果糖含量变化不明显,成熟期蔗糖含量明显提高,但蔗糖∕(葡萄糖+果糖)比值小于1,酸性转化酶(AI)活性随着蔗糖含量的升高而降低,与蔗糖含量呈显著负相关;156和B-3-1-1果实中蔗糖在成熟期快速积累,蔗糖∕(葡萄糖+果糖)比值大于1。研究结果可为甜瓜种质资源创新利用及培育高品质甜瓜提供理论基础。Six different melon inbred lines were used as materials,theβ-carotene content,carotenoid content,the expression of genes related to carotenoid metabolism,the content of glucose,fructose,sucrose and the sucrose metabolism related enzyme activities at different developmental periods were determined.The results showed that the carotenoid content in melon fruit was closely related to the flesh color.With the extension of fruit developmental period,theβ-carotene content and carotenoid content in the fruits of inbred lines 156 and 12B-5-2 increased at first and then decreased,those of B-3-1-1 increased,and those of 162 and 159 decreased.PSY1 gene was highly expressed at the early stage of fruit development in 5 melon varieties with different flesh colors,and gradually decreased with the extension of fruit development.The expression trend of PSY2,LCY-b and ZDS genes in inbred lines 162 and 159 was consistent with PSY1 gene,but the expression trend was opposite in inbred lines wm-6,12B-5-2 and B-3-1-1.PSY2,LCY-b and ZDS genes were expressed most strongly in wm-6 with highest carotenoid content at maturity stage and the weakest expression in the 162,which had the lowest carotenoid content.The correlation between sugar components and content of fruit and flesh color was not obvious.The glucose content in 159 fruits increased gradually with the fruit development,and the sucrose content was the lowest at maturity stage.The fructose content in wm-6,12B-5-2 and 162 fruits did not change significantly.The sucrose content increased significantly at maturity stage,but the sucrose∕(glucose+fructose)ratio was less than 1.The activity of acid invertase(AI)decreased with the increase of sucrose content,and was negatively correlated with sucrose content.In 156 and B-3-1-1 fruits,sucrose accumulated rapidly at maturity stage,and the ratio of sucrose∕(glucose+fructose)was greater than 1.This study could provide a theoretical basis for the innovative utilization of melon germplasm resources and the cultivation of high quality melon
