摘 要:目的探讨自身免疫性肝炎(AIH)小鼠肝组织中的膜联蛋白A1(AnxA1)表达情况及意义。方法选取正常C57BL/6雄性小鼠16只,随机分为正常组和模型组,各8只。模型组通过尾静脉注射刀豆蛋白A造模,正常组通过尾静脉注射等量的生理盐水。48 h后处死小鼠,取肝组织进行苏木精-伊红染色观察组织病理学变化,比较两组小鼠血清ALT、AST水平,以及肝组织AnxA1原位表达情况、肝组织AnxA1 mRNA及蛋白表达水平。分析AnxA1 mRNA及蛋白表达水平与血清ALT、AST水平的相关性。结果模型组血清ALT、AST水平均高于正常组(均P<0.05)。模型组小鼠肝脏充血肿大,镜下可见明显炎症改变。模型组肝组织存在AnxA1的表达,且mRNA和蛋白的相对表达量均高于正常组(均P<0.05)。模型组肝组织AnxA1 mRNA和蛋白的相对表达量均与血清ALT、AST水平呈正相关(均P<0.05)。结论AIH小鼠肝组织AnxA1表达上调,其可能在AIH发生、发展中起到重要的作用。Objective To investigate the expression and significance of annexin A1(AnxA1)in liver tissues of autoimmune hepatitis(AIH)mice.Methods Sixteen normal male C57BL/6 mice were selected,and were randomly divided into normal group and model group,with 8 mice in each group.The model group was modeled by tail vein injection of concanavalin A,and the normal group received tail vein injection of isometric normal saline.After 48 hours,all mice were sacrificed,and liver tissues were removed for observation on histopathological changes after hematoxylin-eosin staining.The two groups were compared in terms of serum ALT and AST levels,as well as the in situ expression of AnxA1 in liver tissues and the expression levels of AnxA1 mRNA and protein in liver tissues.The correlation of AnxA1 mRNA or protein expression level with serum ALT or AST level was analyzed.Results The model group had higher serum ALT and AST levels than the normal group(all P<0.05).The livers of mice in the model group exhibited congestion and edema,and obvious inflammatory changes were observed under microscope.AnxA1 was expressed in the liver tissues of the model group,and the relative expression of AnxA1 mRNA and protein was higher in the model group than in the normal group(all P<0.05).In the model group,the relative expression of AnxA1 mRNA and protein in liver tissues positively correlated with serum ALT or AST level(all P<0.05).Conclusion AnxA1 is highly expressed in liver tissues of AIH mice,and may play an important role in the occurrence and development of AIH.
