摘  要:目的探究B、T淋巴细胞弱化因子(B-and T-lymphocyte attenuator,BTLA)在宫颈鳞癌组织中的表达、临床意义及对Hela细胞生物学行为的影响。方法免疫组化法检测宫颈炎症、鳞状上皮内病变及鳞癌组织中BTLA的表达;使用siRNA干扰Hela细胞内BTLA的表达,并通过CCK-8、Transwell、划痕实验检测其对Hela细胞增殖、侵袭、迁移能力的影响。结果BTLA在宫颈炎症、鳞状上皮内病变及鳞癌组织中的表达水平呈上升趋势(P=0.000),且与宫颈鳞癌的临床分期、肌层浸润深度、淋巴结转移情况等呈正相关(P<0.05);siRNA成功抑制BTLA表达,CCK-8、Transwell及划痕实验显示,siRNA干扰组细胞的增殖、侵袭及迁移能力显著降低(P<0.05)。结论BTLA在宫颈鳞癌组织中呈现高表达,其高表达与宫颈鳞癌的发生、发展相关;靶向抑制BTLA显著下调Hela细胞的增殖、侵袭及迁移能力,BTLA有望成为宫颈鳞癌基因治疗的新靶点。Objective To explore the expression and clinical significance of B and T lymphocyte weakening factor(B-and T-lymphocyte attenuator,BTLA)in cervical squamous cell carcinoma and its effect on the biological behavior of Hela cells.Methods Immunohistochemical method was used to detect the expression of BTLA in cervical inflammation,squamous intraepithelial lesion and squamous cell carcinoma.siRNA was used to interfere with the expression of BTLA in Hela cells,and CCK-8,Transwell and scratch test were used to detect the effect of BTLA on the proliferation,invasion and migration of Hela cells.Results The expression of BTLA in cervical inflammation,squamous intraepithelial lesion and squamous cell carcinoma showed an upward trend(P=0.000),and was positively correlated with clinical stage,depth of myometrial invasion and lymph node metastasis of cervical squamous cell carcinoma(P<0.05).siRNA successfully inhibited the expression of BTLA,CCK-8,Transwell and scratch test showed that the ability of cell proliferation,invasion and migration decreased significantly in siRNA interference group(P<0.05).Conclusion BTLA is highly expressed in cervical squamous cell carcinoma,and its high expression is related to the occurrence and development of cervical squamous cell carcinoma.Targeted inhibition of BTLA significantly down-regulates the ability of proliferation,invasion and migration of Hela cells.BTLA is expected to become a new target for gene therapy of cervical squamous cell carcinoma.
