摘 要:目的研究槲皮素对血小板激活及高血压小鼠缺血性脑卒中的影响。方法C57BL/6小鼠采用两肾一夹(2K1C)建立高血压模型,随机分为假手术组、模型组及低、高剂量檞皮素(30、50 mg·kg^(-1))组。模型建成后,采用线栓法构建小鼠脑梗死模型,1 h后再灌注,同时ip给药,假手术组和模型组予等体积生理盐水,每天1次,直到处死小鼠。脑梗死后24 h,提取各组小鼠血小板检测血小板功能和炎性因子的释放,检测各组小鼠尾出血时间及神经功能损伤、死亡率、脑梗体积和脑血栓形态。结果与对照组比较,高血压组小鼠血小板活化显著增强,小鼠脑卒中加重。槲皮素能显著抑制脑卒中小鼠血小板的活化,减少炎性因子的释放,缩小脑梗体积,降低神经评分。高剂量槲皮素给药的保护作用更强,能显著降低高血压合并脑卒中小鼠的死亡率。且槲皮素对小鼠尾出血时间无明显影响。结论槲皮素能通过抑制高血压合并脑卒中小鼠血小板的活化,减少血小板炎性因子的释放,降低脑血栓中血小板堆积的致密程度和覆盖率,降低高血压合并脑卒中小鼠的脑梗死体积和神经评分,增加小鼠存活率,且无明显的出血风险。OBJECTIVE To investigate effect of quercetin(QE)on platelet activation and cerebral infarction in hypertensive mice.METHODS Two kidneys and one clip(2 K1 C)were used to construct a hypertensive model.C57 BL/6 mice randomly divided into sham operation group,model group,low-dose QE(30 mg·kg^(-1))group and high-dose QE(50 mg·kg^(-1))group.After the model was successfully constructed,all mice were constructed cerebral infarction model(t MCao).Mice were reperfused after occlusion for 1 h.They were treated with drugs by subcutaneous injection at the same time as reperfusion,with mice of sham operation group and model group were given an equal volume of normal saline.The drugs were maintained once a day until the mice were sacrificed.24 h after cerebral infarction,we extracted platelets from each group to detect platelet function and the release of inflammatory factors.The tail bleeding time,neurological damage,mortality,cerebral infarction volume and cerebral thrombosis morphology of mice were detected.RESULTS In model group,platelet activation was significantly enhanced,and the degree of cerebral infarction increased.Quercetin administration significantly inhibited platelet activation,reduced the release of inflammatory factors,the volume of cerebral infarction,and the nerve score.The protective effect of high-dose quercetin was stronger,which could significantly reduce the mortality of mice with hypertension and cerebral infarction.And quercetin does not increase tail bleeding in mice.CONCLUSION Quercetin could inhibit platelet activation,reduce the release of platelet inflammatory factors,the density and coverage of platelet accumulation in cerebral thrombosis,reduce the volume of cerebral infarction and nerve score in hypertensive mice with cerebral infarction,and increase the survival rate of mice,without obvious risk of bleeding.
