摘 要:目的探讨多聚赖氨酸溶液对兔同种异体骨进行表面改性使其具有良好的细胞亲和力与最低细胞毒性的最佳浓度。方法使用浓度为100 mg/mL、30 mg/mL、1 mg/mL、0.1 mg/mL、0.025 mg/mL和0.01 mg/mL的多聚赖氨酸溶液,采取冻干法,分别对兔同种异体骨的微孔进行表面改性。根据多聚赖氨酸浓度,将兔同种异体骨分为100 mg/mL组、30 mg/mL组、1 mg/mL组、0.1 mg/mL组、0.025 mg/mL组和0.01 mg/mL组,同时,将未行表面改性的同种异体骨设为空白对照组。将兔骨髓间充质干细胞(BMSCs)分别种植在各组同种异体骨内,并向成骨方向进行诱导和增殖。采用噻唑蓝(MTT)比色试验检测BMSCs在各组同种异体骨微孔表面的黏附、增殖情况,同时检测各组经诱导培养后的细胞表达碱性磷酸酶(ALP)的活性。结果在经不同浓度多聚赖氨酸溶液表面改性的同种异体骨中,空白对照组和0.01 mg/mL组BMSCs增殖及其表达ALP的活性明显小于其他各组(P<0.05),而其他各组之间差异无统计学意义(P>0.05)。结论从经济、有效方面考虑,多聚赖氨酸溶液对同种异体骨进行表面改性的最佳浓度为0.025 mg/mL,该浓度可使同种异体骨具有良好的细胞亲和力与最低的细胞毒性。Objective To investigate the best concentration of polylysine solution for surface modification of rabbit allograft bone with good cell affinity and minimum cytotoxicity.Methods The polylysine solution with concentrations of 100 mg/mL,30 mg/mL,1 mg/mL,0.1 mg/mL,0.025 mg/mL and 0.01 mg/mL were used to performed the surface modification of the micropore in rabbit allograft bone by freeze-drying method respectively.According to the concentration of polylysine solution,rabbit allograft bone were divided into the 100 mg/mL group,the 30 mg/mL group,the 1 mg/mL group,the 0.1 mg/mL group,the 0.025 mg/mL group and the 0.01 mg/mL group.In addition,the allograft bone without surface modification was set as the blank control groop.Rabbit bone marrow mesenchymal stem cells(BMSCs)were implanted on the allograft bone of each group,and were induced and proliferated in the osteogenic direction.The adhesion and proliferation of BMSCs on the surface of the allograft bone micropore were investigated by the methyl thiazolyl tetrazolium(MTT)colorimetric test.And the activity of alkaline phosphatase(ALP)expressed by the cells in all groups after induction and culture was detected.Results In the allograft bone which was surface-modified by polylysine solution with different concentrations,the proliferation and the activity expressing ALP of BMSCs in the blank control group and the 0.01 mg/mL group were significantly less than those in the other groups(P<0.05),but there was no significant difference in the other groups(P>0.05).Conclusion In terms of economy and efficiency,the best concentration of polylysine solution for surface modification of allograft bone is 0.025 mg/mL,which can make allograft bone have good cell affinity and minimum cytotoxicity.
