摘 要:目的探讨胸腺基质淋巴生成素对大鼠皮肤损伤修复作用及免疫机制。方法52只SPF级雌性大鼠随机分为观察组与对照组,每组各26只。观察组采用特制致伤器建立大鼠皮肤损伤模型。免疫组化法检测大鼠皮肤组织中胸腺基质淋巴生成素表达情况;酶联免疫吸附法测定血清胸腺基质淋巴生成素、干扰素γ(IFN-γ)、白细胞介素4(IL-4)、免疫球蛋白E(IgE)水平。结果胸腺基质淋巴生成素阳性表达于细胞核,在细胞膜和细胞浆中未见阳性着色;观察组平均光密度为(82.60±9.34),明显高于对照组的(50.38±3.91),差异有统计学意义(P<0.01)。观察组大鼠血清胸腺基质淋巴生成素、IL-4、IgE水平明显高于对照组,IFN-γ水平明显低于对照组,差异有统计学意义(P<0.01)。Pearson相关性分析显示,观察组大鼠血清胸腺基质淋巴生成素与IFN-γ呈负相关,与IL-4、IgE呈正相关。结论胸腺基质淋巴生成素在大鼠皮肤损伤组织中高表达,且与免疫炎症因子明显相关,可能参与皮肤损伤的修复过程。Objective To investigate the repair action and immune mechanism of thymic stromal lymphopoietin on skin injury in rats.Methods A total of 52 SPF female rats were randomly divided into the observation group(n=26)and the control group(n=26).In the observation group,the rat skin injury model was established with a special injury device.Immunohistochemical method was used to detect the expression of thymic stromal lymphopoietin in rat skin tissue.The levels of thymic stromal lymphopoietin,interferonγ(IFN-γ),interleukin 4(IL-4)and immunoglobulin E(IgE)in serum were measured by enzyme-linked immunosorbent assay.Results Thymic stromal lymphopoietin was positively expressed in the nucleus,but no positive staining was found in the cell membrane and cytoplasm.The average optical density in the observation group was(82.60±9.34),significantly higher than(50.38±3.91)in the control group,with statistically significant difference(P<0.01).The levels of serum thymic stromal lymphopoietin,IL-4 and IgE in the observation group were significantly higher than those in the control group,while the level of IFN-γin the observation group was significantly lower than that in the control group(P<0.01).Pearson correlation analysis showed that serum thymic stromal lymphopoietin in the observation group was negatively correlated with IFN-γand positively correlated with IL-4 and IgE.Conclusion Thymic stromal lymphopoietin is highly expressed in the injured skin tissue of rats,and is remarkably related to immune inflammatory factors,which may be involved in the repair process of skin injury.
