Effect of Early Acupuncture on Neurological Functions and Cleaved Caspases-3 Expression in Rats with Traumatic Brain Injury

摘  要:目的观察针刺对颅脑损伤(TBI)大鼠脑组织Cleaved Caspases-3表达、脑组织病理形态和运动、感觉等神经功能的影响。方法将40只大鼠随机分为空白组、假手术组、模型组、针刺组,各10只。模型组和针刺组应用PCI3000打击器撞击硬脑膜制备TBI模型;假手术组仅开颅暴露硬脑膜,不进行打击;空白组不做处理。造模后即对针刺组进行针刺治疗,每日1次,干预7 d。在治疗第2、4、7天应用改良神经功能评分(mNSS)和Rotarod test检测大鼠运动、感觉等神经功能损伤程度。第7天治疗结束后利用HE染色法观察脑组织病理形态改变,Western blotting法检测脑组织Cleaved Caspases-3蛋白表达。结果治疗第2、4、7天时与假手术组相比,模型组mNSS评分明显升高(均P <0.01),Rotarod test评分显著下降(P <0.01,P <0.01,P <0.05)。治疗第4、7天时,与模型组相比,电针组大鼠mNSS评分降低(均P <0.05),Rotarod test评分升高(P <0.05或P <0.01)。HE染色显示针刺组可观察到正常神经元,组织疏松和紊乱轻于模型组,整体情况好于模型组。模型组大鼠Cleaved Caspases-3蛋白表达量显著高于假手术组(P <0.01),针刺组表达量低于模型组(P <0.01)。结论针刺可以通过抑制脑组织内Cleaved Caspases-3的表达,减轻细胞凋亡达到修复脑组织和保护神经功能的疗效。Objective:To observe the effect of acupuncture on the expression of cleaved Caspases-3 in brain tissue,the pathological morphology of brain tissue and the neurological functions of motor and sensory function in rats with traumatic brain injury(TBI).Methods:A total of 40 rats were randomly divided into the blank group,sham group,model group and acupuncture group,10 rats in each group.In the model group and acupuncture group,TBI model was made by striking the cerebral dura mater with PCI3000;the dura mater was only exposed without striking in the sham group;the blank group underwent nothing.The acupuncture group was treated with acupuncture once a day for 7 days after operation.On the 2 nd,4 th and 7 th day of treatment,mNSS score and Rotarod test were performed to detect the degree of motor and sensory neurological impairment of the rats.After the 7 th day,HE staining was used to observe the pathological changes of brain tissue,and Western blotting was used to detect the protein expression of cleaved Caspases-3.Results:On the 2 nd,4 th and 7 th day of treatment,the mNSS score of the model group was significantly higher than that of the sham operation group(all P<0.01),and the Rotarod test score was significantly decreased(P<0.01,P<0.01,P<0.05).On the 4 th and 7 th day of treatment,compared with the model group,the m NSS score of EA group was decreased(all P<0.05),and the Rotarod test score was increased(P<0.05,P<0.01).HE staining showed that normal neurons could be observed in the acupuncture group,the tissue loose and disorder were lighter than the model group,and the overall situation was better than the model group.The expression of cleaved Caspases-3 protein in the model group was significantly higher than that in the sham operation group(P<0.01),and the relative protein expression in the acupuncture group was lower than that in the model group(P<0.01).Conclusion:Acupuncture can repair brain tissue and protect nerve function by inhibiting the expression of cleaved Caspases-3 and reducing apoptosis.
