异补骨脂素促进大鼠骨髓间充质干细胞向骨细胞成分化并抑制其向脂肪细胞分化

摘　　要：目的探究异补骨脂素对于促进大鼠骨髓间充质干细胞向成骨分化及抑制其向脂肪细胞分化的实际作用。方法从动物实验有限公司处购买10只实验用清洁级Wistar大鼠,杀死后取双侧股骨和胫骨,获得骨髓,并提取充质干细胞。由研究组自行制备成骨细胞诱导溶液以及成脂细胞抑制诱导溶液后。其中成骨细胞诱导实验,将异补骨脂素浓度分为0.1、1、10μmol/L共3组进行,作为补充试验的补骨脂素组,浓度为10μmol/L;成脂细胞抑制诱导实验中的异补骨脂素浓度为1μmol/L。结果异补骨脂素对大鼠骨髓间充质干细胞的CFU-f、ALP阳性CFU-f的促进作用十分明显,且随着异补骨脂素添加计量的增多,单位时间内CFU-f的形成效率也呈现显著的上升趋势。当浓度达到1μmol/L时,作用程度达到最大值,再度提升浓度,CFU-f的形成效率并未出现显著提升。与未接受任何定向引导的常规干细胞相比,实验组干细胞脂肪滴的数量呈显著下降的趋势,且脂肪细胞体积明显减少。结论异补骨脂素对促进大鼠骨髓间充质干细胞向成骨细胞分化并抑制其向脂肪细胞分化的作用较为明显。Objective Objective to explore the effect of isopsoralen on promoting osteogenic differentiation and inhibiting adipocyte differentiation of rat bone marrow mesenchymal stem cells.Methods Ten experimental clean grade Wistar rats were purchased from animal experiment Co.,Ltd.after being killed,bilateral femurs and tibias were taken to obtain bone marrow,and mesenchymal stem cells were extracted.The study group prepared osteoblast induction solution and adipocyte inhibition induction solution.In the osteoblast induction experiment,isopsoralen concentration was divided into three groups:0.1μmol/L,1μmol/L and 10μmol/L.The psoralen group was 10μmol/L,and the isopsoralen concentration in the inhibition and induction experiment of osteoblasts was 1μmol/L.Results Isopsoralen significantly promoted the CFU-F and ALP positive CFU-F of rat bone marrow mesenchymal stem cells,and with the increase of isopsoralen dosage,the formation efficiency of CFU-F per unit time also showed a significant upward trend.When the concentration of CFU-F reached 1μmol/L,the degree of action reached the maximum.When the concentration was increased again,the formation efficiency of CFU-F did not increase significantly.Compared with the conventional stem cells without any directional guidance,the number of adipose droplets and the volume of adipocytes in the experimental group decreased significantly.Conclusion Isopsoralen can promote the differentiation of rat bone marrow mesenchymal stem cells into osteoblasts and inhibit their differentiation into adipocytes.
