脑小血管病性认知障碍与外周血血浆外泌体浓度的相关性

摘　　要：目的探讨血浆外泌体在脑小血管病(cerebral small vessel disease,CSVD)性认知障碍患者中的表达及意义。方法30例CSVD性认知障碍患者作为实验组,另选取30例健康志愿者作为对照组,使用蒙特利尔认知评估量表(MoCA)评估认知功能。利用血浆外泌体试剂盒提取受试者血浆外泌体,应用电镜及粒径分析验证外泌体,使用BCA方法测得各组血浆外泌体浓度水平。结果观察组MoCA总评分及注意、语言、执行、记忆、计算力、定向力、视结构技能评分均显著低于健康组,差异有统计学意义(P<0.05);实验组与对照组血浆外泌体浓度分别为(4.62±1.37)、(5.83±1.77)mg/L,两组比较差异显著(P<0.05)。结论CSVD性认知障碍患者血浆外泌体浓度降低,血浆外泌体有望成为CSVD性认知障碍潜在的诊断标志物。Objective To investigate the expression and significance of plasma exosomes in patients with cognitive impairment caused by cerebral small vessel disease(CSVD).Methods 30 cases of patients with CSVD cognitive impairment were selected as the experimental group,and another 30 cases of healthy volunteers as the control group.Montreal Cognitive Assessment Scale(MoCA)was used to assess cognitive function.Plasma exosomes were extracted by the plasma exosomes kit.Electron microscopy and particle size analysis verified exosomes.The concentration of plasma exosomes was measured by BCA method.Results The total score of MOCA and the scores of attention,language,execution,memory,calculation,orientation and visual structure skills in the observation group were significantly lower than those in the healthy group(P<0.05).The concentrations of plasma exosomes in the experimental group and the control group were(4.62±1.37)mg/L and(5.83±1.77)mg/L,respectively,with significant difference between the two groups(P<0.05).Conclusion The concentration of plasma exosomes decreased in CSVD patients with cognitive impairment,plasma exosomes may be a potential diagnostic marker for cognitive impairment in csvd patients.
