摘  要:目的分析流感病毒检测中细胞培养法与实时荧光定量聚合酶链反应(PCR)法的应用效果。方法选取2018年10月~2020年10月我中心604例疑似流感样本为研究对象,分别通过细胞培养法和实时荧光定量PCR法进行检测,比较检测结果。结果实时荧光定量PCR法阳性率为26.49%,高于细胞培养法的12.58%,差异有统计学意义(P<0.05);实时荧光定量PCR阳性样本分离率为37.50%,高于阴性样本分离率的3.60%,差异有统计学意义(P<0.05);以细胞培养法为金标准,实时荧光定量PCR法检测灵敏度为90.79%,特异度为98.42%。结论实时荧光定量PCR法检测流感病毒的应用价值较高,具有检测速度快、灵敏度高、特异性强等优势,可用于应急检测、常规检测等。细胞培养能够获取毒株,有利于进一步对病毒抗原性、基因特性以及耐药性的研究。Objective To analyze the application effect of cell culture method and real-time fluorescent quantitative polymerase chain reaction(PCR)method in influenza virus detection.Methods From October 2018 to October 2020,604 samples of suspected influenza in our center were selected as the research object,and the detection results were compared by cell culture method and real-time fluorescent quantitative PCR method.Results The positive rate of real-time fluorescent quantitative PCR method was 26.49%,which was higher than 12.58%of cell culture method,the difference was statistically significant(P<0.05);The separation rate of real-time fluorescent quantitative PCR positive samples was 37.50%,which was higher than 3.60%of negative samples,the difference was statistically significant(P<0.05);Taking cell culture method as the gold standard,the detection sensitivity of real-time fluorescent quantitative PCR method is 90.79%,and the specificity was 98.42%.Conclusion The real-time fluorescent quantitative PCR method has high application value in detecting influenza virus.It has the advantages of fast detection speed,high sensitivity,and strong specificity.It can be used for emergency detection and routine detection.Cell culture can obtain virus strains,which is conducive to further research on virus antigenicity,genetic characteristics and drug resistance.
