摘  要:目的观察不同佐剂与抗原表位融合蛋白配伍后对季节性流感疫苗在小鼠体内免疫原性的影响。方法流感病毒抗原表位融合蛋白(FP)分别辅以氢氧化铝、聚肌胞(PolyIC)+氢氧化铝以及阳离子佐剂免疫小鼠,再以四价流感病毒裂解疫苗(QIV)加强免疫,通过检测血凝抑制(hemagglutination inhibition,HI)效价和血凝素(hemagglutinin,HA)IgG抗体评价免疫原性。结果佐剂和融合蛋白本身并不能产生针对任何型别的HI抗体;QIV加强免疫后,阳离子佐剂配伍组显著提高了QIV同源株H1N1和H3N2型HI效价或阳转率,且提高了非同源株H3N2型HA IgG抗体水平。结论阳离子佐剂辅助流感病毒抗原表位融合蛋白免疫小鼠后,可显著增强季节性流感疫苗甲型毒株在小鼠体内的免疫原性。Objective To observe the effect of different adjuvants combined with epitope fusion protein(FP) on immunogenicity of seasonal influenza vaccines in mice. Methods Mice were primed with influenza virus epitope FP supplemented with aluminum hydroxide,PolyIC + aluminum hydroxide and cationic adjuvant respectively,then boosted with quadrivalent influenza virus split vaccine(QIV). The immunogenicity was evaluated by determination of hemagglutination inhibition(HI)titer and hemagglutinin(HA)IgG antibody. Results Adjuvant and FP produced no HI antibody of any types alone. However,after booster immunization with QIV,the HI titer or positive conversion rate against homologous strains of H1N1 and H3N2 types as well as the HA IgG antibody level against non-homologous strains of H3N2 type in the mice immunized with FP combined with cationic adjuvant increased significantly. Conclusion Influenza virus epitope FP combined with cationic adjuvant significantly enhanced the immunogenicity of seasonal influenza vaccine strain A in mice.
