和胃理气方对肝郁型功能性消化不良大鼠Ghrelin表达的影响

摘　　要：目的研究肝郁型功能性消化不良大鼠胃排空率以及胃和下丘脑组织Ghrelin蛋白的表达变化,以探究和胃理气方对功能性消化不良干预的可能机制。方法选取40只健康Wistar大鼠,雌雄各半,体质量(200±20)g,随机分为空白组(10只)及模型组(30只)。空白组常规饲养,其余大鼠通过夹尾刺激法建立肝郁型FD大鼠模型,造模14 d。造模结束后按照随机数字表分为模型组、吗丁啉组、和胃理气方组(每组10只)。期间监测大鼠进食以及毛发、行为等一般状况。实验结束后,测量各组大鼠胃排空率,并通过Western blot法测定胃组织以及下丘脑组织Ghrelin蛋白表达。结果模型组胃排空率明显低于和胃理气方组、吗丁啉组(P<0.05);与模型组相比,和胃理气方组和吗丁啉组的胃及下丘脑Ghrelin蛋白表达增加(P<0.05)。结论和胃理气方可能通过上调Ghrelin蛋白表达来有效干预功能性消化不良。Objective:To study the gastric emptying rate and the expression changes of Ghrelin protein in stomach and hypothalamus in rats with liver-stagnation functional dyspepsia in order to explore the possible mechanism of Hewei Liqi decoction on functional dyspepsia.Methods:40 healthy Wistar rats were selected,half male and half male,and weighed(200±20)g.They were randomly divided into a blank group(10)and a model group(30).The blank group was bred routinely and the rest of the rats were used to create a liver depression type FD rat model by the tail-clamping method,and the model was established for 14 days.After modeling,the rats were divided into model group,domperidone group and Hewei Liqi Decoction group(10 each group)according to the random number table.During this period,the general conditions of the rats'food intake,hair,behavior,etc.were monitored.After the experiment,the gastric emptying rate of rats in each group was measured and the expression of Ghrelin protein in gastric tissue and hypothalamic tissue was determined by Western blot.Results:The gastric emptying rate of the model group was significantly lower than that of the Hewei Liqi Decoction group and the domperidone group(P<0.05).Compared with that of the model group,the gastric and hypothalamic Ghrelin protein expression in the Hewei Liqi Decotion group and the domperidone group increased(P<0.05).Conclusion:Hewei Liqi Decoction may effectively interfere with functional dyspepsia by up-regulating the expression of Ghrelin protein.
