摘 要:目的:探讨大孔树脂纯化漆姑草总黄酮工艺。方法:比较6种不同型号大孔吸附树脂对漆姑草中总黄酮的吸附和解吸效果,筛选出纯化漆姑草总黄酮的最佳树脂,并对其吸附和解吸条件进行考察和优化,应用单因素实验,确定最佳纯化工艺。结果:AB-8树脂具有最佳吸附和解吸特性,最佳工艺为上样液浓度1.076 mg/mL,pH为5.7,上样体积6 BV,以3 BV 70%乙醇在流速2 BV/h洗脱,得黄酮的质量分数为75.50%。结论:AB-8树脂用于纯化漆姑草总黄酮,工艺稳定可靠。Objective To explore the the technique of purifying the total flavonoids of Herba Saginae Japonicae(FHS)by macroporous resin.Method The adsorption and desorption effects of 6 different types of macroporous adsorption resins on FHS was compared to screen out the best resin for purifying FHS,and investigated and optimized its adsorption and desorption conditions.the best optimization process was determined by application single factor experiment.Results AB-8 resin had the best adsorption and desorption characteristics.The best process was the sample concentration 1.076 mg/mL,pH 5.7,sample volume 6 BV,3 BV 70%ethanol at sampling velocity of 2 BV/h eluting,and the mass fraction of flavonoids was 75.50%.Conclusion The AB-8 resin is used to purify FHS and the technique is stable and reliable.
