摘 要:目的基于近红外光谱分析技术结合偏最小二乘法建立快速测定灯盏花素注射液含量的方法。方法选取2个厂家生产的27批灯盏花素注射液及实验室自制的54个样品为建模样本,首先以液体附件采用透反射法采集样品的近红外光谱,同时以标准方法测定含量,进而利用化学计量学方法辅助建立模型。结果基于内部交叉验证构建模型。测定样本的含量范围为0.956~4.15 mg/ml;建模谱段为7502~5446 cm-1和4602~4247 cm^(-1);预处理方法选择一阶导结合矢量归一化;Rank值为7;交叉验证的均方根误差为0.24%,决定系数为95.9%。结论所建定量模型准确性、耐用性良好,可对灯盏花素注射液进行快速定量分析,同时为近红外技术应用于中药分析开拓了思路。Objective To establish a quick method for determination of Breviscapine Injections based on near infrared trans-reflection spectroscopy technology combined with partial least square algorithm.Methods 27 batches of Breviscapine Injections from 2 different manufactures and 54 self-made samples in laboratory were used as samples for modeling.Firstly,the near infrared spectra of the samples were collected by the transmission-reflection method with the liquid accessories,and the contents were determined by the standard method;then the chemometric method was used to assist the establishment of the model.Results The model was built based on cross validation.The content range of the samples determined was 0.956-4.15 mg/ml.The modeling wavelength ranges of the spectra were 7502-5446 cm-1&4602~4247 cm-1.The pretreatment method was first order derivative combined with vector normalization.The Rank value was 7,and the values of RMSECV and R2 in the cross validation were 0.24%and 95.9%,respectively.Conclusion The quantitative model built has good accuracy and durability which can analyze quantitatively Breviscapine Injections quickly.Meanwhile,it opened up a new way for the application of near infrared technology in analysis of traditional Chinese medicine.
