摘 要:目的探讨NaHoF_(4)@CeO_(2)纳米颗粒(nanoparticles,NPs)CT、MRI双模态显像的可行性及其对胰腺癌细胞(Panc-1)的体外放疗增敏效果。方法合成并表征NaHoF_(4)@CeO_(2)纳米颗粒,并测定其粒径大小及主要元素物质含量。分别用MTT及动物实验测定NPs在体内外的毒性作用,CT及MRI成像仪评估NPs体外CT及MRI成像效果,MTT及细胞克隆实验验证单纯NPs组,单纯放疗(radiation therapy,RI)组及RI+NPs组对Panc-1放疗增敏效果。结果成功制备出粒径大小均匀、化学成分稳定的纳米复合材料NaHoF_(4)@CeO_(2)。NPs在体外无明显的细胞毒性作用,体外实验表明NPs具有良好的CT及MRI双模态成像作用。此外,体外放疗MTT实验及细胞克隆实验表明该材料对胰腺癌细胞有放疗增敏效果。结论NaHoF_(4)@CeO_(2) NPs可同时进行CT、MRI双模态显像,通过体外细胞学实验表明,该纳米粒子具有对Panc-1细胞放疗增敏效果。Objective To study the synthesis of NaHoF_(4)@CeO_(2)Nanoparticles(NPs),and the feasibility of CT and MRI dual-mode imaging,and the radiotherapy sensitization effect to the pancreatic cancer cells(Panc-1)in vitro.Methods We synthesized NaHoF_(4)@CeO_(2)NPs and measured the particle size and main contents of elements.We used MTT and animal experiments to determine the toxic effects of NPs in vitro and in vivo and evaluated the effect of the NPs in vitro CT imaging and MRI by the CT and MRI imager.MTT and cell cloning experiments were performed to verify the effect of radiotherapy on Panc-1 in the pure NPs group,Radiation therapy(RI)group,and RI+NPs group.Results The nanocomposite material NaHoF_(4)@CeO_(2)with uniform particle size and stable chemical composition was successfully prepared.NPs had no obvious cytotoxic effect in vitro.In vitro experiments showed that NPs had good dual-modality imaging effects of CT and MRI.In addition,in vitro radiotherapy MTT experiments and cell cloning experiments showed that the NPs had a radiosensitization effect on pancreatic cancer cells.Conclusion NaHoF_(4)@CeO_(2)NPs can simultaneously perform as CT and MRI dual-mode imaging,and it has the effect of radiotherapy sensitizing on Panc-1 cells through in vitro cytological experiments.
