摘  要:通过转录组测序技术(RNA-seq)对尖孢镰刀菌(Fusariumoxysporum)侵染条件下河南烤烟主栽品种‘中烟100’根系的差异表达基因进行了分析,并对侵染后根系和叶片4种内源激素含量变化、根系抗氧化酶类活性、酚类化合物次生代谢相关酶类活性和总抗氧化能力、超氧阴离子清除率、脯氨酸和丙二醛含量变化进行了研究。结果表明:镰刀菌侵染烟株矮小,主根和侧根的数量和体积显著降低,平均直径增加。烤烟根系基因表达发生显著变化,差异基因的功能主要涉及不同生物学过程、细胞组分和分子功能等几个方面,新陈代谢相关的Unigene所占比例最高。侵染后烤烟根系和叶片的IAA、GA3、ZA和ABA内源激素含量均有不同程度升高。SOD等抗氧化酶类活性、酚类化合物次生代谢酶类活性、谷胱甘肽氧化还原反应酶类活性、总抗氧化能力和超氧阴离子清除率、脯氨酸等渗透调节物质含量均显著升高,APX活性显著降低。抗氧化能力、代谢酶类以及保护性物质含量的升高有利于烟株阻挡病原菌入侵,从而降低病害对烤烟造成的危害,提高烟株抗病性。The differentially expressed genes in the roots of Zhongyan 100 infected by Fusarium oxysporum were analyzed by RNA-seq technique.The changes of four endogenous hormones in roots and leaves,the antioxidant enzyme activity in roots,the phenolic compound secondary metabolism related enzyme activity,the total antioxidant ability,the superoxide anion clearance rate and the contents of T-AOC,PRO and MDA were studied.The results showed that the infected tobacco growed lower.The number and volume of tap roots and lateral roots reduced significantly,and the average diameter increased.The expression of genes in flue-cured tobacco roots changed significantly.The functions of differentially expressed genes mainly involved in different biological processes,cell components and molecular functions,and the metabolism-related unigene had the highest proportion.The contents of IAA,GA3,ZA and ABA in infected roots and leaves increased at different degrees.The activity of antioxidant enzymes such as SOD,phenolic compound secondary metabolism related enzymes and glutathione redox reaction related enzymes,total antioxidant ability,superoxide anion clearance rate and osmotic adjustment substances such as proline increased significantly,and the activity of APX decreased significantly.The increase of antioxidant ability and contents of metabolic enzymes and protective substances was benefit for tobacco roots to block the invasion of pathogenic bacteria,thereby reducing the damage caused by diseases and improving the disease resistance of tobacco plants.
