摘 要:目的观察牛蒡苷元(Arc-G)对血小板体外聚集作用的影响,并初步探讨其作用机制。方法制备小鼠洗涤血小板,分为模型组、替罗非班组(0.5 g/L)、Arc-G低剂量组(3.125μmol/L)及Arc-G高剂量组(12.5μmol/L),采用比浊法测定凝血酶(250 U/L)、胶原(0.5 mg/L)及二磷酸腺苷(ADP,0.25μmol/L)诱导的血小板聚集率;将洗涤血小板分为模型组、PGE 1组(前列腺素E 1,1μmol/L)或替罗非班组(0.5 g/L)、Arc-G低剂量组、Arc-G高剂量组,采用ELISA实验方法测定凝血酶诱导聚集下血小板内环磷酸腺苷(cAMP)、血栓素B 2(TXB 2)的含量。结果与模型组比较,Arc-G高剂量能明显抑制凝血酶、ADP诱导的血小板聚集,差异有统计学意义(P<0.05),但不能抑制胶原诱导的血小板聚集;与模型组比较,Arc-G低剂量、Arc-G高剂量可明显升高血小板内cAMP的水平、降低TXB 2的含量,差异有统计学意义(P<0.05)。结论Arc-G可明显抑制凝血酶和ADP诱导的血小板聚集,其作用可能与升高血小板内cAMP的水平、降低血小板TXB 2的水平有关。Objective To observe the effect of arctigenin(Arc-G)on platelet aggregation in vitro and discuss the mechanism.Methods Preparation of mouse washing platelets were divided into model group,tirofiban group(0.5 g/L),Arc-G low dose group(3.125μmol/L),the Arc-G high dose(12.5μmol/L).The platelet aggregation rate induced by thrombin(250 U/L),collagen(0.5 mg/L)and adenosine diphosphate(ADP,0.25μmol/L)was determined by turbidimetry.The washed platelets were divided into model group,PGE 1 group(prostaglandin E 1,1μmol/L)or tiroffei group(0.5 g/L),Arc-G low-dose group and high-dose group.The contents of cyclic adenosine monophosphate(cAMP)and thromboxane B 2(TXB 2)in platelets under thrombin-induced aggregation were determined by ELISA experimental method.Results Arc-G high dose significantly inhibited thrombin and ADP induced platelet aggregation compared with the model group,but did not inhibit collagen-induced platelet aggregation.The difference was statistically significant(P<0.05).Arc-G low-dose and high-dose could significantly increase the level of platelet cAMP and decrease the content of TXB 2 compared with the model group,and the difference was statistically significant(P<0.05).Conclusion Arc-G can significantly inhibit thrombin and ADP induced platelet aggregation,and its effect may be related to increasing the level of platelet cAMP and reducing the level of platelet TXB 2.
