摘  要:目的探讨反复丙泊酚麻醉对成年鼠海马突触可塑性和学习记忆的影响。方法45只SD雄性大鼠按随机数字表法分为对照组(n=15)、丙泊酚1组(n=15)与丙泊酚2组(n=15),3组分别在特制的麻醉箱中给予腹腔注射0.9%氯化钠溶液、25 mg/kg与50 mg/kg丙泊酚,1次/d,持续10 d。检测与记录实验第5、10天3组大鼠的120 s内穿越原平台位置的次数与逃避潜伏期;实验第5、10天采用酶联免疫吸附试验(ELISA)检测3组大鼠白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)水平;实验第10天采用TUNEL法检测海马组织CA1区细胞凋亡水平;采用蛋白质印迹(Western blotting)法检测大鼠的海马组织Caspases-3、Bax、海马突触后致密物(PSD-95)、突触素(SYN)蛋白相对表达水平。结果丙泊酚1组与丙泊酚2组实验第5、10天的穿越平台次数少于对照组,逃避潜伏期多于对照组,差异均有统计学意义(P<0.05),丙泊酚2组穿越平台次数少于丙泊酚1组,逃避潜伏期多于丙泊酚1组,差异均有统计学意义(P<0.05)。丙泊酚1组与丙泊酚2组实验第5、10天的血清IL-1β、TNF-α水平高于对照组,差异均有统计学意义(P<0.05);丙泊酚2组血清IL-1β、TNF-α水平高于丙泊酚1组,差异均有统计学意义(P<0.05)。丙泊酚1组与丙泊酚2组实验第10天的海马组织细胞凋亡指数、Caspases-3、Bax蛋白相对表达水平高于对照组,PSD-95、SYN蛋白相对表达水平低于对照组,差异均有统计学意义(P<0.05);丙泊酚2组较丙泊酚1组细胞凋亡指数和以上蛋白表达水平变化更显著,差异均有统计学意义(P<0.05)。结论反复丙泊酚麻醉可促进成年鼠海马组织Caspases-3和Bax蛋白的表达,降低PSD-95、SYN蛋白表达,促进神经元细胞凋亡,诱发机体出现炎症反应,从而影响大鼠的突触可塑性和学习记忆能力。Objective To investigate the effects of repeated propofol anesthesia on synaptic plasticity and learning and memory of adult rat hippocampus.Methods A total of 45 male SD rats were divided into control group(n=15),propofol 1 group(n=15)and propofol 2 group(n=15)according to the random number table method.The three groups were given intraperitoneal injection of 0.9%sodium chloride solution,25 mg/kg and 50 mg/kg propofol in a special anesthesia box,once a day for 10 days.On the 5th and 10th day of the experiment,the number of crossing the original platform position and the escape latency within 120 s of the 3 groups of rats were detected and recorded;on the 5th and 10th day of the experiment,the level of interleukin-1β(IL-1β)and tumors necrosis factor-α(TNF-α)of the 3 groups of rats was detected by enzyme-linked immunosorbent assay(ELISA);on the 10th day of the experiment,the level of apoptosis in the CA1 area of hippocampus was detected by TUNEL method;the relative expression levels of Caspases-3,Bax,postsynaptic density-95(PSD-95),and synaptophysin(SYN)proteins in the hippocampus of rats were detected by Western blotting.Results The crossing platforms in the propofol 1 group and propofol 2 group were fewer than the control group on the 5th and 10th day,and the escape latency were more than those of the control group,the differences were statistically significant(P<0.05).The crossing platforms in the propofol 2 group were fewer than the propofol 1 group on the 5th and 10th day,and the escape latency were more than those of the propofol 1 group,the differences were statistically significant(P<0.05).The levels of serum IL-1βand TNF-αin the propofol group 1 and propofol group 2 were higher than those in the control group on the 5th and 10th day,and levels of serum IL-1βand TNF-αin propofol group 2 were higher than those of the propofol group 1,and the differences were statistically significant(P<0.05).The apoptosis index,Caspases-3 and Bax protein relative expression levels in the CA1 area of the hippocampus
