摘  要:【目的】对贵州省火龙果溃疡病的病原进行鉴定及生物学特性研究,为贵州火龙果溃疡病的防治及病原菌的碳氮源代谢研究奠定理论基础。【方法】采用组织分离法、形态学观察以及分子生物学等方法对贵州火龙果溃疡病病原进行分离鉴定,并采用菌丝生长速率法及Biolog表型芯片技术研究病原菌生物学特性。【结果】贵州火龙果溃疡病病菌为新暗色柱节孢(Neoscytalidium dimidiatum);病原菌适宜生长温度为20~35℃,最佳生长温度为28℃,最适pH为6~7;光照对菌丝生长有抑制作用,短时间的紫外光照射对病原菌生长有促进作用。火龙果溃疡病菌对PMl、PM2微孔板中190种碳源物质利用率为18.42%,对PM3微孔板中95种氮源物质的利用率为52.63%。【结论】贵州火龙果溃疡病病菌为新暗色柱节孢(Neoscytalidium dimidiatum),病原菌较耐高温,生长环境偏酸性。【Objective】The pathogen identification and research on biological characteristics of pitaya canker in Guizhou Province are conducted to lay the theoretical basis for prevention and control of Guizhou pitaya canker and research on carbon and nitrogen metabolism of pathogen.【Method】The pathogen of Guizhou pitaya canker is isolated and identified by using tissue isolation method,morphological observation and molecular biology,and then the biological characteristics of the pathogen are studied by employing mycelium growth rate and Biolog phenotype MicroArray technology.【Result】The pathogen of Guizhou pitaya canker is identified as Neoscytalidium dimidiatum.The suitable growth temperature,optimum growth temperature and optimum pH for the pathogen are 20—35℃,28℃and 6—7.Illumination has inhibition effect on mycelium growth,while short-time UV irradiation promotes the growth of pathogen.The utilization rate of N.dimidiatum to 190 carbon-source materials in PM1 and PM2 microplate is 18.42%,and the utilization rate to 95 nitrogen-source materials in PM3 microplate is 52.63%.【Conclusion】The pathogen of Guizhou pitaya canker is identified as N.dimidiatum.The pathogen is resistant to high temperature,and grows at acidic environment.
