摘  要:目的:为进一步开发利用药食同源植物资源,本研究以马齿苋醇提物为研究对象,探索其醇提物的抑菌、美白、抗敏、降糖、降脂活性。方法:测定马齿苋醇提物对常见5种供试菌的最小抑菌浓度和最小杀菌浓度,评价其抑菌活性;测定马齿苋醇取物对透明质酸酶、酪氨酸酶及α-葡萄糖苷酶活性抑制率的影响,评价其美白、抗敏、降糖功效;利用MTT法检测不同浓度的马齿苋醇提物对氧化型低密度脂蛋白(ox-LDL)诱导的Raw 264.7细胞增殖活力影响及降脂作用;结果:马齿苋醇提物对5种常见供试菌均有抑制效果,其中对白色念珠菌抑制效果最佳;当马齿苋醇提物浓度为20 mg·mL-1时,对透明质酸酶的体外抑制率为89.66%;浓度为20 mg·mL-1时,其对酪氨酸酶的抑制率高达76.71%,抑制效果是同等浓度熊果苷的1.25倍;200 mg·mL-1马齿苋醇提物对α-葡萄糖苷酶抑制率达49%;MTT法检测结果显示在100~400μg·mL-1浓度内,对Raw 264.7细胞增殖活力无显著影响,且能显著降低细胞内总胆固醇(TC)及游离胆固醇(FC)含量。结论:综合试验表明马齿苋可以作为一种安全无刺激,且具有保健功效的药食两用植物原料,被开发成具有降糖、降脂功效的保健品,以及具有美白、抗敏作用的护肤品和天然抑菌剂。该研究结果为进一步开发利用马齿苋资源和提高经济效益提供了一定的理论基础。Objective:In order to further develop and utilize medicinal and edible plant resources,this study took the ethanol extract of purslane as the research object to exp-lore its antibacterial,whitening,anti-allergy,hypoglycemic and lipid-lowering activities.Methods:The minimum inhibitory concentration and minimum bactericidal concentration of the ethanol extract from purslane against 5 kinds of common tested bacteria were determined,and its antibacterial activity was evaluated.The effects of purslane extract on the activity inhibition rates of hyaluronidase,tyrosinase andα-glucosidase were determined,and the whitening,anti-sensitization and hypoglycemic effects of purslane extract were evaluated.MTT assay was used to detect the effects of different concentrations of ethanol extract of purslane on the proliferation and lipid-lowering activity of Raw 264.7 cells induced by oxidized low-density Lipoprotein(ox-LDL).Results:The ethanol extract of purslane had inhibitory effect on 5 kinds of common tested bacteria,among which Candida albicans had the best inhibitory effect.When the concentration of the ethanol extract of purslane was 20 mg·mL-1,the inhibition rate of hyaluronidase was 89.66%in vitro.When the concentration was 20 mg·mL-1,the inhibition rate of tyrosinase was as high as 76.71%,which was 1.25 times that of arbutin at the same concentration.The inhibitory rate of 200 mg·mL-1 purslane extract onα-glucosidase was 49%.The results of MTT assay showed that the proliferation activity of Raw 264.7 cells was not significantly affected at the concentration of 100~400μg·mL-1,and the contents of intracellular total cholesterol(TC)and free cholesterol(FC)were significantly reduced.Conclusion:The comprehensive experiment shows that purslane can be used as a safe and non-stimulating plant material with health care effects,and can be developed into health care products with the effects of lowering blood sugar and lipid,skin care products with whitening and anti-sensitization effects and natural bacteriostatic agents.T
