HPLC法测定中药袋中4种抗氧剂的含量及其迁移

摘　　要：目的:建立中药袋中抗氧剂1010、330、1076、168的含量测定方法。并用此方法监测4种抗氧剂的迁移量。方法:采用Waters ZORBAX eclipse XDB C_(18)色谱柱(250 mm×4.6 mm,5μm),以甲醇为流动相,流速1.0 ml/min,柱温25℃,检测波长274 nm。装入90℃的模拟浸泡液(水、pH 3.5缓冲液、pH 8.0缓冲液)模拟灌装,分别监测0、3和7天的迁移量。结果:抗氧剂1010、330、1076和168分别在0.00508~0.06102 mg/ml(r=0.9989)、0.05058~0.60690 mg/ml(r=0.9999)、0.05000~0.60000 mg/ml(r=1.0000)、0.05298~0.63570 mg/ml(r=0.9997)范围内呈良好的线性关系。平均加样回收率分别为82.4%、84.5%、91.6%和95.7%,RSD分别为0.5%、1.1%、1.5%和1.3%。8批中药袋中均未检出抗氧剂330,检测到抗氧剂1010、1076、168的中药袋在迁移试验中均有不同程度的迁移。结论:该方法简单准确、重复性好,可用于中药袋中抗氧剂的质量控制。Objective:To establish a method for the determination of antioxidant 1010,330,1076,168 in bag of traditional Chinese medicine.The migration of four antioxidants was monitored by this method.Methods:The Water ZORBAX eclipse XDB C_(18) column(250 mm×4.6 mm,5μm)was adopted with the mobile phase of methanol at a flow rate of 1.0 ml/min.The column temperature was at 25℃,the detection wavelengths was at 274 nm.The simulated immersion solution(water,pH 3.5 buffer,pH 8.0 buffer)was filled at 90℃,and the migration was monitored for 0,3 and 7 days respectively.Results:The calibration curves of antioxidant 1010,330,1076,168 were linear within the ranges of 0.00508~0.06102 mg/ml(r=0.9989),0.05058~0.60690 mg/ml(r=0.9999),0.05000~0.60000 mg/ml(r=1.0000),0.05298~0.63570 mg/ml(r=0.9997),respectively;the average recoveries of the antioxidants were 82.4%,84.5%,91.6%and 95.7%with the RSDs of 0.5%,1.1%,1.5%and 1.3%,respectively.Antioxidant 330 was not found in 8 batches of Chinese herbal medicine bags,antioxidant 1010,1076 and 168 were detected to have different migration degree in the migration experiment.Conclusion:This method is simple,accurate,with good repeatability and can be used for quality control of antioxidants in traditional Chinese medicine bags.
