摘  要:目的观察活动期SLE患者IL-38表达水平变化,并探讨IL-38对IκB激酶复合物(IκK)的调控作用。方法采用实时荧光定量聚合酶链反应(PCR)检测IL-38、IκB激酶复合物基因表达水平;ELISA检测血清IL-38水平;蛋白印迹法检测IκKα/β表达水平;采用t检验或Mann-Whitney秩和检验。结果①活动期SLE患者IL-38 mRNA表达水平(0.36±0.09)显著低于健康对照组(1.00±0.17),差异有统计学意义(Z=-4.07,P<0.01);活动期SLE患者IL-38蛋白表达水平(5.86±2.76)显著低于健康对照组(18.48±1.35),差异有统计学意义(Z=-4.76,P<0.05);②活动期SLE患者IκKα和IκKβmRNA表达水平(7.45±0.31)和(6.01±1.51)显著高于健康对照组(1.16±0.04)和(1.16±0.14),差异有统计学意义(Z=-4.67,P<0.05;Z=-4.37,P<0.01);且活动期SLE患者IL-38 mRNA表达水平分别与IκKα和IκKβmRNA表达水平呈负相关(r=-0.78,P<0.05;r=-0.83,P<0.05);③活动期SLE患者非磷酸化IκKα/β蛋白表达水平(2.38±0.03)高于健康对照组(1.00±0.04),差异有统计学意义(Z=-1.96,P<0.05);磷酸化IκKα/β蛋白表达水平(1.90±0.03)高于健康对照组(1.00±0.08),差异有统计学意义(Z=-1.99,P<0.05);④在体外实验中,活动期SLE患者外周血中加入IL-38后,IκKα和IκKβmRNA表达水平(1.70±0.12)和(2.52±0.10)较未处理活动期SLE患者明显下降(2.56±0.28)和(3.82±0.38),差异有统计学意义(Z=-1.96,P<0.05;Z=-1.37,P<0.05);⑤在体外实验中,活动期SLE患者外周血中加入IL-38后,非磷酸化IκKα/β蛋白表达水平(1.54±0.06)较未处理活动期SLE患者组明显下降(2.93±0.08),差异有统计学意义(Z=-2.09,P<0.05);磷酸化IκKα/β蛋白表达水平(0.970±0.012)较未处理活动期SLE患者组明显下降(1.572±0.051),差异有统计学意义(Z=-2.24,P<0.05)。结论活动期SLE患者外周血IL-38表达降低,并且与IκKα和IκKβ水平呈负相关;在体外实验中,IL-38能够降低IκB激酶复合物的表达,推测在SLE患者体内IL-38水平下降,造成IκB激酶复合物的Objective To observe the expression of interleukin(IL)-38 in patients with active systemic lupus erythematosus(SLE),and to explore the regulatory effect of IL-38 on IκB kinase complex.Methods The gene expression levels of IL-38 and IκB kinase complex were detected by real-time fluorescence quantitative polymerase chain reaction(PCR).Serum IL-38 levels were detected by enzyme-linked immunosorbent assay(ELISA).The expression of IκB-kinase complex(IκK)α/βand phospho IκKα/βprotein weredetermined by Western blotting analysis.Statistical analysis was conducted with or Mann-Whitney rank test.Results①The expression level of IL-38 mRNA in active SLE patients(0.36±0.09)was significantly lower than that in the normal control group(1.00±0.17)(Z=-4.07,P<0.01);the expression level of IL-38 protein in active SLE patients(5.86±2.76)significantly reduced as compared with normal control group(18.48±1.35)(Z=-4.76,P<0.05).②The expression level of IκKαmRNA(7.45±0.31)and IκKβmRNA(6.01±1.51)in active SLE patients was significantly higher than that in normal control group(1.16±0.04)and(1.16±0.14)(Z=-4.67,P<0.05;Z=-4.37,P<0.01),and the expression level of IL-38 mRNA was negatively correlated with IκKαmRNA and IκKβmRNA in active SLE patients(r=-0.78,P<0.05;r=-0.83,P<0.05).③IκKα/βand phosphorylated IκKα/βprotein expression in active SLE patients(2.38±0.03)and(1.90±0.03)increased significantly compared with healthy controls(1.00±0.04)and(1.00±0.08)(Z=-1.96,P<0.05;Z=-1.99,P<0.05).④In vitro experiment,IL-38 caused a significant decrease in the expression level of IκKαmRNA(1.70±0.12)and IκKβmRNA(2.52±0.10)from active SLE patientscompared with untreated cells(2.56±0.28)and(3.82±0.38)(Z=-1.96,P<0.05;Z=-1.37,P<0.05).⑤In vitro experiment,IL-38 caused a significant decrease in the expression level of IκKα/β(1.54±0.06)and phosphoryl-ated IκKα/β(0.970±0.012)protein expression in active SLE patients compared with untreated cells(2.93±0.08)and(1.572±0.051)(P<0.05).Conclusion The de-crease o
