摘  要:目的比较寻常性银屑病患者皮损与正常对照角质形成细胞(keratinocytes,KCs)的糖酵解和线粒体有氧呼吸水平,揭示患者能量代谢水平是否异常,进一步探讨银屑病可能的发病机制。方法分离、培养寻常性银屑病患者皮损与健康人皮肤KCs,采用Seahorse XF24细胞能量代谢分析仪检测细胞的糖酵解和线粒体有氧呼吸水平。结果与正常对照相比,寻常性银屑病患者皮损KCs的整体糖酵解水平出现异常升高,糖酵解代谢发生紊乱;寻常性银屑病患者皮损KCs的线粒体有氧呼吸水平与正常对照相比差异无统计学意义(P>0.05)。结论寻常性银屑病患者皮损的KCs能量代谢水平出现紊乱,过度依赖于糖酵解代谢,快速产生大量的ATP,从而为其过度增殖提供能量和原料支持。Objective To compare the glycolysis and mitochondrial aerobic respiration of keratinocytes(KCs)in psoriasis vulgaris patients with normal controls,to reveal whether the energy metabolism level of patients is abnormal,and to further explore the possible pathogenesis of psoriasis.Methods KCs were isolated and cultured from skin lesions of psoriasis vulgaris patients and healthy people.Glycolysis and mitochondrial aerobic respiration were measured by Seahorse XF24 cell energy metabolism analyzer.Results Compared with the normal control group,the overall glycolysis level of KCs in psoriasis vulgaris patients was increased abnormally and glycolysis metabolism was disordered.The mitochondrial aerobic respiration level of KCs in psoriasis vulgaris patients was not significantly different from that in normal control group.Conclusion The energy metabolism of KCs which depends excessively on glycolysis metabolism in psoriasis vulgaris patients is disordered,and produces a large amount of ATP rapidly,thereby providing energy and raw material support for its excessive proliferation.
