白花蛇舌草内生真菌BY1不同提取部位对人胃癌细胞SGC-7901增殖和凋亡的作用研究

摘　　要：目的:研究白花蛇舌草内生真菌BY1乙酸乙酯提取部位、氯仿提取部位、甲醇提取部位对人胃癌细胞SGC-7901(SGC-7901细胞)增殖和凋亡的作用。方法:体外培养SGC-7901细胞,采用MTT法测定BY1不同提取部位对SGC-7901细胞干预48 h的增殖抑制作用,采用Hoechst 33258染色法观察SGC-7901细胞的凋亡变化。结果:BY1不同提取部位对SGC-7901细胞均具有抑制作用,由强到弱依次为氯仿部位、乙酸乙酯部位和甲醇部位,对SGC-7901细胞半数抑制浓度(IC50)分别为177.37±11.53μg/ml、380.53±40.07μg/ml和1077.80±40.17μg/ml。细胞凋亡形态可观察到核固缩、核碎片和聚集等,药物干预48 h后凋亡率增加呈剂量-效应关系。结论:白花蛇舌草内生真菌BY1对SGC-7901细胞具有良好的抑制作用,并可诱导其凋亡。Objective:Toinvestigate the effects of different extracts from endophytic fungus BY 1 of Hedyotis diffusa Willd on proliferation and apoptosis of human gastric cancer SGC7901 cells(SGC7901 cells).Methods:SGC7901 cells were cultured in vitro,the proliferation of SGC7901 cells was detected by MTT assay after treating with different extracts from BY1 for 48 h,and the apoptosis morphology of SGC7901 cells was determined by Hoechst 33258 fluores⁃cent staining.Results:Different extracts from BY1 had inhibitory effect on SGC7901 cells,three extracts from strong to weak were Chloroform fraction,Ethyl acetate fraction and Methanol fraction,the 50%inhibitory concentration(IC50)of SGC7901 cells at 48 h was 177.37±11.53μg/ml,380.53±40.07μg/ml and 1077.80±40.17μg/ml.Apoptosis morpholo⁃gy as nuclear condensation,fragmentation and aggregation was observed,and the apoptosis ratio of SGC7901 cells was increased at 48 h in dose-effect manner.Conclusion:Different extracts from BY1 had good inhibition to SGC7901 cells,and could induce apoptosis of SGC7901 cells.
