摘  要:目的探讨促血管生成素1(Ang-1)基因转染至骨髓间充质干细胞(BMSCs)后对高氧诱导的新生鼠视神经损伤模型的协同保护作用。方法 30只出生后3 d的健康小鼠,随机分为高氧盐水组、空白对照组、Ang-1+BMSCs高氧组,每组10只。三组小鼠均置于安静、室温和湿度适宜的环境下进行饲养,造模期间可自由摄食和饮水。比较三组小鼠有髓神经纤维数目、直径、轴索直径及髓鞘厚度。结果空白对照组、Ang-1+BMSCs高氧组轴索直径分别为(0.72±0.10)、(0.70±0.12)μm,显著长于高氧盐水组的(0.60±0.10)μm,差异具有统计学意义(P<0.05);三组轴索直径比较差异具有统计学意义(P<0.05)。空白对照组、Ang-1+BMSCs高氧组有髓神经纤维直径分别为(1.06±0.10)、(0.99±0.13)μm,长于高氧盐水组的(0.90±0.16)μm,差异具有统计学意义(P<0.05);三组有髓神经纤维直径比较差异具有统计学意义(P<0.05)。高氧盐水组、Ang-1+BMSCs高氧组有髓神经纤维数目低于空白对照组, Ang-1+BMSCs高氧组神经纤维数量明显高于高氧盐水组,差异具有统计学意义(P<0.05);三组有髓神经纤维数量比较差异具有统计学意义(P<0.05)。空白对照组、 Ang-1+BMSCs高氧组髓鞘厚度大于高氧盐水组,差异具有统计学意义(P<0.05);三组髓鞘厚度比较差异具有统计学意义(P<0.05)。结论高氧诱导新生鼠视神经损伤后进行Ang-1转染至BMSCs,可显著改善损伤的视神经超微结构。Objective To discuss the synergistic protective effect of Angiopoietin 1(Ang-1)gene transfected into bone manrrow mesenchymal stem cells(BMSCs)on hyperoxia induced optic nerve injury in neonatal rats.Methods A total of 30 healthy mice 3 d after birth were randomly divided into hyperoxia saline group,blank control group,Ang-1+BMSCs hyperoxia group,with 10 rats in each group.The mice of three groups were raised in a quiet,room temperature and humidity environment,and they were free to eat and drink during the model building.The number and diameter of myelinated nerve fibers,axonal diameter and myelin thickness in the three groups were compared.Results The axonal diameter of blank control group and Ang1+BMSCs hyperoxia group were(0.72±0.10)and(0.70±0.12)μm,which were significantly longer than(0.60±0.10)μm of hyperoxia saline group,and the difference was statistically significant(P<0.05).There was statistically significant difference in axonal diameter among the three groups(P<0.05).The diameter of myelinated nerve fibers of blank control group and Ang-1+BMSCs hyperoxia group were(1.06±0.10)and(0.99±0.13)μm,which were longer than(0.90±0.16)μm of hyperoxia saline group,and the difference was statistically significant(P<0.05).There was statistically significant difference in diameter of myelinated nerve fibers among three groups(P<0.05).The numbers of myelinated nerve fibers of hyperoxia saline group and Ang-1+BMSCs hyperoxia group were lower than that of the control group,and Ang-1+BMSCs hyperoxia group was obviously higher than that of hyperoxia saline group.All the difference was statistically significant(P<0.05).There was statistically significant difference in number of myelinated nerve fibers among the three groups(P<0.05).The myelin thickness of blank control group and Ang-1+BMSCs hyperoxia group was greater than that of hyperoxia saline group,and the difference was statistically significant(P<0.05).There was statistically significant difference in myelin thickness among the three groups(P<0.05).Conclu
