摘 要:细胞异质性是多细胞生物体中普遍存在的现象。通过分离单细胞,并对其进行转录组分析,有助于准确而深入地理解细胞分化、信号转导等生物学过程及相关的基因调控网络。目前动物细胞的相关研究已取得显著进展,而植物由于存在细胞壁,显著增加了细胞分离的难度及基因表达数据的准确度,限制了植物单细胞转录组测序的研究进展。本研究综述了几种有较广阔应用前景的植物单细胞分离技术,重点介绍了根细胞和保卫细胞两类典型细胞的单细胞转录组学研究进展,并讨论了植物单细胞转录组学研究中存在的问题及未来展望。Cellular heterogeneity is a common phenomenon in multicellular organisms. By isolating single cells and conducting transcriptome analysis, it is helpful to accurately and deeply understand biological processes such as cell differentiation, signal transduction and related gene regulatory networks. At present, significant progress has been made in the studies of animal cells, while the existence of cell walls in plants significantly increases the difficulty of cell separation and the accuracy of gene expression data, which limits the research progress of single-cell transcriptome sequencing in plants. In this summarize, several techniques for single cell separation in plants with promising applications are reviewed, with emphasis on the research progress of single cell transcriptomics in root cells and guard cells, and the existing problems and future prospects of single cell transcriptomics in plants are discussed.
